目的:建立HPLC法测定双黄连粉针剂中黄芩苷血浓度并研究其在家兔体内药物动力学。方法:采用Hypersil C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-水-磷酸(25:75:0.2);流速:0.8 ml·min-1;检测波长:275 nm;柱温:30℃;进样量20μl。结果:线性范围为0.5~595.3 mg·L-1,平均回收率为(93.2±7.1)%,日内、日间RSD均小于10.0%(n=5),最低检测浓度为0.1 mg·L-1。黄芩苷在家兔体内主要药动学参数:t1/2α=6.7 min,t1/2β=118.8 min,V(c)=0.0866 L·kg-1,AUC0-t= 8980.1 mg·min·L-1,AUC0-∞=9204.4 mg·min·L-1,MRT0-t=14.2 min,MRT0-∞=17.9 min,CL(s)=0.0079 L·min-1·kg-1。结论:该法适于黄芩苷及其制剂的血浓度测定。
Objective: To establish HPLC method for determination of baicalin blood concentration in Shuanghuanglian injection and study its pharmacokinetics in rabbits. Methods: A Hypersil C18 column (250 mm×4.6 mm, 5 μm) was used; mobile phase: acetonitrile-water-phosphoric acid (25:75:0.2); flow rate: 0.8 ml·min-1; detection wavelength : 275 nm; column temperature: 30°C; injection volume 20 μl. Results: The linear range was 0.5-595.3 mg·L-1, the average recovery was (93.2±7.1)%, and the intra- and inter-day RSD was less than 10.0% (n=5). The lowest detection concentration was 0.1 mg·L-1. The main pharmacokinetic parameters of baicalin in rabbits: t1/2α=6.7 min, t1/2β=118.8 min, V(c)=0.0866 L·kg-1, AUC0-t=8980. 1 mg·min·L-1, AUC0-∞=9204.4 mg·min·L-1, MRT0-t=14.2 min, MRT0-∞=17.9 min, CL(s)=0.0079 L·min-1·kg-1. Conclusion: This method is suitable for determination of blood concentration of baicalin and its preparations.
