目的了解弓形虫感染对糖尿病鼠肾脏功能的影响。方法取OLETF鼠20只,分为实验组和对照组(各10只)。于30周龄时实验组每鼠感染106个弓形虫速殖子(0.1ml),对照组注射等量生理盐水。分别于30、32、34、36、38、40周龄时取血,采用全自动生化分析仪测定肌酐(Scr)、尿素氮(BUN)、尿酸(UA)、肌酸激酶(CK)、总胆固醇(CH)、甘油三脂(TG),采用免疫浊度法测定纤维蛋白原含量;收集鼠尿,采用双缩脲法测24h尿蛋白,判断弓形虫感染对糖尿病鼠肾脏功能的影响。结果弓形虫感染组OLETF鼠34、36、38、40、周龄时的血肌酐与对照组比较差异有统计学意义(t34=3.3750,t36=4.6512,t38=6.2637,t40=6.6747,均P<0.01),尿素氨差异有统计学意义(t34=6.8198,t36=4.0757,t38=4.2556,t40=4.5844,P<0.05或P<0.01);36、38、40周龄时的血纤维蛋白原与对照组比较差异有统计学意义(t36=3.0710,t38=4.2111,t40=3.8726,P<0.05或P<0.01);38周龄时的24h尿蛋白与对照组比较差异有统计学意义(t38=2.2483,t40=5.1645,P<0.05或P<0.01);尿酸、肌酸激酶、总胆固醇及甘油三脂两组间差异无统计学意义(P>0.05)。结论弓形虫感染加重糖尿病OLETF鼠肾功能损害,采取有效措施防止弓形虫感染可保护糖尿病鼠肾脏功能。
Objective To understand the effect of Toxoplasma gondii infection on renal function in diabetic rats. Methods Twenty OLETF mice were divided into experimental group and control group (10 in each). At 30 weeks of age, 106 Toxoplasma tachyzoites (0.1 ml) were infected in each experimental group, while the control group was injected with the same amount of normal saline. Blood samples were taken at 30, 32, 34, 36, 38 and 40 weeks of age respectively. The levels of creatinine, blood urea nitrogen, urine uric acid and creatine kinase were measured by automatic biochemical analyzer. Cholesterol (CH) and triglyceride (TG) were measured by enzyme-linked immunosorbent assay (ELISA). The content of fibrinogen was determined by immunoturbidimetry. Urine was collected and 24h urinary protein was measured by biuret method to determine the effect of Toxoplasma gondii infection on renal function in diabetic rats. Results The levels of serum creatinine in the Toxoplasma gondii infection group were significantly higher than those in the control group (t34 = 3.3750, t36 = 4.6512, t38 = 6.2637, t40 = 6.6747, P < 0.01). There was a significant difference in urea ammonia (t34 = 6.8198, t36 = 4.0757, t38 = 4.2556, t40 = 4.5844, P <0.05 or P <0.01) (T36 = 3.0710, t38 = 4.2111, t40 = 3.8726, P <0.05 or P <0.01). There was a significant difference in 24h urine protein between the control group and the control group (t38 = 2.2483, t40 = 5.1645, P <0.05 or P <0.01). There was no significant difference in uric acid, creatine kinase, total cholesterol and triglyceride between the two groups (P> 0.05). Conclusion Toxoplasma gondii infection aggravates renal dysfunction in diabetic OLETF mice and effective measures to prevent Toxoplasma gondii infection can protect renal function of diabetic mice.
