目的探讨B细胞特异的莫洛尼白血病病毒插入位点1(Bmi-1)基因在子宫内膜癌组织中的表达及意义。方法采用免疫组化SP法检测57例子宫内膜癌和18例正常子宫内膜(子宫肌瘤子宫切除患者)石蜡标本中的Bmi-1蛋白表达;采用RT-PCR法检测子宫内膜癌组织及正常子宫内膜组织(各10例)Bmi-1 mRNA的表达。结果 57例子宫内膜癌中,Bmi-1蛋白阳性29例(50.9%),18例正常子宫内膜中无Bmi-1蛋白阳性表达;病理学G3级Bmi-1蛋白阳性表达高于G1级(66.7%vs.30.4%)(P<0.05)。子宫内膜癌中Bmi-1mRNA电泳条带灰度比高于正常子宫内膜组织[(1.0375±0.0910)vs.(0.5546±0.0698)](P<0.05)。结论癌基因Bmi-1可能与子宫内膜癌的发生、发展及恶性程度有关。
Objective To investigate the expression and significance of B cell-specific Moloney leukemia virus insertion site 1 (Bmi-1) gene in endometrial carcinoma. Methods Immunohistochemical SP method was used to detect the expression of Bmi-1 protein in 57 cases of endometrial carcinoma and 18 cases of normal endometrium (hysteromyoma hysterectomy) paraffin specimens; RT-PCR was used to detect the expression of endometrial carcinoma And normal endometrium (10 cases each) Bmi-1 mRNA expression. Results In 57 cases of endometrial carcinoma, Bmi-1 protein was positive in 29 cases (50.9%) and 18 cases of normal endometrium without Bmi-1 protein expression. The pathological G3-Bmi-1 protein expression was higher than that in G1 (66.7% vs.30.4%) (P <0.05). The gray scale ratio of Bmi-1mRNA bands in endometrial carcinoma was higher than that in normal endometrial tissues [(1.0375 ± 0.0910) vs. (0.5546 ± 0.0698)] (P <0.05). Conclusion The oncogene Bmi-1 may be related to the occurrence, development and malignancy of endometrial carcinoma.
