目的探讨三氧化二砷与顺铂联合对体外培养卵巢癌细胞株HO8910细胞增殖、细胞周期调控及细胞凋亡影响。方法用三氧化二砷与顺铂联合处理HO8910细胞,采用MTT法检测药物对细胞的抑制作用,倒置显微镜观察细胞形态学改变;琼脂糖凝胶电泳观察DNA降解,以及应用流式细胞仪观察细胞凋亡过程中细胞周期的变化。结果在三氧化二砷与顺铂联合作用下,HO8910细胞呈凋亡改变,DNA琼脂糖凝胶电泳呈典型的凋亡特征。细胞凋亡的同时,细胞周期发生特定的改变。结论三氧化二砷与顺铂联合能抑制卵巢癌细胞增殖,能诱导卵巢癌细胞凋亡。
Objective To investigate the effects of arsenic trioxide combined with cisplatin on the proliferation, cell cycle and apoptosis of ovarian cancer cell line HO8910 in vitro. Methods HO8910 cells were treated with arsenic trioxide combined with cisplatin. MTT assay was used to detect the inhibitory effect of the drugs on the cells. The morphological changes of the cells were observed by inverted microscope. The DNA degradation was observed by agarose gel electrophoresis. The apoptosis of cells was observed by flow cytometry Changes in the cell cycle. Results Apoptosis of HO8910 cells was observed under arsenic trioxide combined with cisplatin. DNA agarose gel electrophoresis showed typical apoptotic characteristics. Apoptosis at the same time, the cell cycle occurs a specific change. Conclusion Combination of arsenic trioxide and cisplatin can inhibit the proliferation of ovarian cancer cells and induce the apoptosis of ovarian cancer cells.
