转化生长因子β(transforming growth factors-β,TGF-β)是一类具有多种生物学活性的细胞因子,参与调节细胞的增殖、分化、发育和凋亡等多种生命活动。TGF-β对肿瘤的作用是极其复杂的。在空间方面,TGF-β信号通路对肿瘤细胞自身的作用在肿瘤抑制和肿瘤发展机制中有重要意义,又能通过影响肿瘤的微环境(如成纤维细胞、免疫细胞和细胞外基质)来促进或抑制肿瘤的发展和转移;在时相方面,TGF-β在肿瘤早期可抑制细胞增殖、启动细胞分化并诱导凋亡,但在肿瘤的进展期则可通过多种机制促进肿瘤的侵袭和转移。TGF-β信号通路的复杂特性对肿瘤机制的研究和肿瘤的靶向治疗提供了全新的机遇和挑战。
