目的 了解大鼠应激性溃疡与胃酸、前列腺素 (PGs)和促肾上腺皮质激素 (ACTH)水平彼此间的关系及其发病机理 ,并进行防治。方法 通过冷束缚法制成大鼠应激性溃疡模型 ,以东莨菪碱、地塞米松、雷尼替丁、洛赛克等药物进行应激性溃疡的防治 ,检测胃液量、pH值及胃粘膜的变化 ,测定PGs和ACTH的水平。结果 正常大鼠饥饿 2 4小时后抽到少量胃液 ;经冷束缚后 ,胃液量增加 ,pH值下降 ,胃粘膜出血 ,血中PGs下降 ,ACTH增加。提示在应激状态下 ,胃粘膜出血与胃液中 pH值下降、PGs含量下降和ACTH含量上升有关。 结论 防治应激性溃疡以升高胃液 pH值能达到目的 ;PGs升高对防治大鼠胃粘膜出血有明显作用 ;洛赛克提高胃液 pH值而不降低PGs的作用 ,可作为防治应激性溃疡的首选药。
Objective To investigate the relationship between stress ulcer and levels of gastric acid, prostaglandin (PGs) and adrenocorticotropic hormone (ACTH) in rats and their pathogenesis and to prevent and cure them. Methods The model of stress ulcer was induced by cold restraint. The rats were treated with scopolamine, dexamethasone, ranitidine and Losec to prevent gastric ulcer. The gastric juice volume, pH and gastric mucosal changes , Determine the level of PGs and ACTH. Results In normal rats, a small amount of gastric juice was drawn after 24 hours of starvation. After cold restraint, gastric juice volume increased, pH value decreased, gastric mucosal hemorrhage, blood PGs decreased and ACTH increased. Prompted in stress state, gastric mucosal bleeding and gastric pH, PGs content decreased and ACTH content increased. Conclusion The prevention and treatment of stress ulcer can increase the pH value of gastric juice to achieve the purpose; PGs increased prevention and treatment of gastric mucosal bleeding has a significant role; Losec increase gastric juice pH without reducing the role of PGs can be used as a control stress The drug of choice for ulcers.
