数十年来,左旋多巴曾为帕金森病(PD)治疗带来革命性的改变,并始终作为PD治疗的金标准享有及其重要的地位。然而,伴随其治疗产生的运动并发症却成为困扰医患的一大难题。近年来,随着对运动并发症机制的研究,越来越多的证据表明持续性多巴胺能刺激(CDS)可在产生良好疗效的同时降低运动并发症的风险,故其被作为PD治疗的新理念备受关注。为此,大量研究致力于开发能形成CDS状态的左旋多巴新制剂,从而探寻出提高疗效与降低运动并发症之间的平衡点。文中就左旋多巴治疗PD的回顾及进展予以介绍。
For decades, levodopa has revolutionized the treatment of Parkinson’s disease (PD) and has always enjoyed its status as the gold standard for PD treatment. However, with its treatment of motor complications have become a major problem plagued doctors and patients. In recent years, with the research on the mechanism of motor complications, there is more and more evidence that sustained dopaminergic stimulation (CDS) can reduce the risk of motor complications while producing good therapeutic effect. Therefore, it has been treated as a new PD Idea of concern. To this end, a large number of studies devoted to the development of a new preparation of levodopa capable of forming a CDS state have led to the discovery of a balance between improved efficacy and reduction of motor complications. In this paper, the review and progress of levodopa treatment of PD be introduced.
