目的探讨宁夏枸杞粗提物对大鼠溃疡性结肠炎的作用。方法采用三硝基苯磺酸(TNBS)-乙醇溶液(含TNBS20~25mg,乙醇浓度30~50%,大鼠TNBS给药剂量80~90mg/kg)建立溃疡性结肠炎大鼠模型,分别给予柳氮磺胺吡啶(SASP)、宁夏枸杞粗提物以及SASP和枸杞粗提物联合用药后,观察各组大鼠一般情况、结肠病变大体形态及病理变化,并检测大鼠血清SOD和结肠组织中MPO活性。结果宁夏枸杞粗提物能明显降低结肠炎大鼠的死亡率,改善一般情况,缩小溃疡面积。枸杞粗提物组、SASP组DAI评分和CMDI评分明显降低,与模型组比较,差异有统计学意义(P<0.05);模型组大鼠血清SOD和结肠MPO活性与正常组比较,均有统计学差异(P<0.05);宁夏枸杞粗提物组大鼠血清SOD和结肠MPO活性与模型组相比,均有统计学差异(P<0.05);与SASP组相比,无统计学差异(P>0.05);联合用药组大鼠血清SOD和结肠MPO活性与粗提物组及SASP组相比,均有统计学差异(P<0.05)。结论宁夏枸杞粗提物具有抗大鼠溃疡性结肠炎的作用,可升高大鼠血清中SOD,降低结肠组织中MPO活性;联合用药抗溃疡性结肠炎作用优于单用宁夏枸杞粗提物和SASP,实验结果为进一步研究宁夏枸杞抗溃疡性结肠炎的作用机制提供了实验依据。
Objective To investigate the effect of Lycium barbarum crude extract on ulcerative colitis in rats. Methods A rat model of ulcerative colitis was established by using TNBS-ethanol solution (containing TNBS 20-25 mg, ethanol concentration 30-50% and TNBS 80-80 mg / kg) Sulfasalazine (SASP), Ningxia Chinese wolfberry crude extract and SASP and medlar crude extract combined with the general situation of rats in each group, the general morphology and pathological changes of colonic pathology, and the detection of serum SOD and colon tissue MPO activity. Results Ningxia Wolfberry crude extract can significantly reduce the mortality of colitis rats to improve the general situation and reduce the ulcer area. The content of DAI and CMDI in SASP group was significantly lower than that in model group (P <0.05), and the activity of SOD and MPO in model group were statistically higher than those in normal group (P <0.05). Compared with the model group, the activities of SOD and MPO in the serum of Ningxia wolfberry crude extract group were significantly different (P <0.05). Compared with the SASP group, there was no significant difference P> 0.05). Compared with the crude extract group and the SASP group, the serum SOD and the colonic MPO activity in the combination group were significantly different (P <0.05). Conclusion The crude extract of Lycium barbarum has anti-ulcerative colitis in rats, which can increase SOD in serum and decrease the activity of MPO in colonic tissue. The combination of anti-ulcerative colitis is better than that of Ningxia medlar SASP, the experimental results provide experimental evidence for further study on the mechanism of Ningxia wolfberry anti-ulcerative colitis.
