目的观察菊粉联合游泳训练对代谢综合征(MS)大鼠骨骼肌胰岛素受体底物1(IRS-1)、葡萄糖转运体子4(GluT4)表达的影响。方法高糖高脂高盐喂养结合腹腔注射STZ建立MS大鼠模型,干预8周后检测各组FBG、HbA_1c及FIns,计算HOMA-IR;HE染色法观察各组肝脏病理变化;免疫印迹及免疫组织化学法检测大鼠骨骼肌IRS-1、GluT4表达。结果与正常饮食对照组比较,模型组FBG、HbA_1c、FIns及HOMA-IR升高(P<0.01),肝脏出现非酒精性脂肪性肝病(NAFLD)病理改变,骨骼肌IRS-1、GluT4表达降低(P<0.01)。与模型组比较,菊粉联合游泳训练组FBG、HbA_1c、FIns及HOMA-IR降低(P<0.01),NAFLD减轻,骨骼肌IRS-1、GluT4表达升高(P<0.01)。结论菊粉联合游泳训练降低了MS大鼠FBG、HbA_1c及FIns水平,减轻了NAFLD病理改变,改善IR,其机制可能与上调骨骼肌IRS-1、GluT4表达有关。
Objective To observe the effects of inulin training on insulin receptor substrate 1 (IRS-1) and glucose transporter 4 (GluT4) expression in skeletal muscle of metabolic syndrome (MS) rats. Methods High glucose, high fat and high salt diet combined with intraperitoneal injection of STZ were used to establish MS rat model. After intervention for 8 weeks, FBG, HbA_1c and FIns in each group were detected to calculate HOMA-IR. Liver pathological changes were observed by HE staining. Western blot and immunofluorescence Histochemical method was used to detect the expression of IRS-1 and GluT4 in skeletal muscle of rats. Results Compared with the normal diet control group, the FBG, HbA 1c, FIns and HOMA-IR in the model group were increased (P <0.01), the pathological changes of NAFLD and the expression of IRS-1 and GluT4 in skeletal muscle were decreased (P <0.01). Compared with model group, FBG, HbA_1c, FIns and HOMA-IR of inulin-co-swimming training group were decreased (P <0.01), NAFLD was reduced, skeletal muscle IRS-1 and GluT4 expression were increased (P <0.01). Conclusions Inulin co-swimming training reduces the levels of FBG, HbA 1c and FIns in MS rats, alleviates the pathological changes of NAFLD and improves IR. The mechanism may be related to up-regulation of IRS-1 and GluT4 expression in skeletal muscle.
