秦皮甲素对角叉菜胶致小鼠足肿胀及炎症的作用

目的:研究秦皮甲素(esculin,ES)对角叉菜胶致小鼠足肿胀及炎症反应的作用及其机制。方法:以角叉菜胶诱发小鼠足跖炎症模型为实验动物模型。末次给药后,热板法检测痛阈值,并检测小鼠足肿胀;取足跖组织制备匀浆液,ELISA法测定足跖组织中炎症因子IL-1β和TNFα水平。结果:与正常对照组比较,角叉菜胶模型组小鼠足肿胀度显著增高(P<0.01),足跖组织中IL-1β和TNFα含量显著增加(P<0.01,P<0.01)。与角叉菜胶模型组比较,ES可显著提高痛阈(P<0.05);ES可显著降低角叉菜胶诱导的足肿胀及炎症因子IL-1β和TNFα水平(P<0.01,P<0.05,P<0.05)。结论:ES对角叉菜胶致小鼠足肿胀具有显著改善作用,机制可能与其抑制炎症因子合成及释放相关。 Objective: To study the effect and mechanism of esculin (ES) on carrageenan-induced foot swelling and inflammation in mice. Methods: Carrageenan-induced mouse plantar inflammation model as experimental animal model. After the last administration, the pain threshold was detected by hot plate method and the foot swelling of mice was measured. The homogenate was taken from the plantar tissue and the levels of inflammatory cytokines IL-1β and TNFα were measured by ELISA. Results: Compared with the normal control group, the carrageenan significantly increased in the carrageenan model group (P <0.01). The content of IL-1β and TNFα in the paw tissue increased significantly (P <0.01, P <0.01). Compared with carrageenan group, ES significantly increased pain threshold (P <0.05); ES significantly decreased carrageenan-induced paw edema and IL-1β and TNFα levels (P <0.01, P <0.05 , P <0.05). Conclusion: ES can significantly improve carrageenan-induced paw edema in mice. The mechanism may be related to its inhibition of the synthesis and release of inflammatory cytokines.
