The authors used affinity column chromatography and radioimmunoassay (RIA) to study the binding characteristics of serum AFP to concanavalin (Con A) in patients with primary hepatocellular carcinoma and secondary liver cancer. A total of 21 patients with primary hepatocellular carcinoma and 8 patients with secondary liver cancer (two groups of patients confirmed by liver biopsy and histological examination) were selected for testing. A 5.0 ml Con A-glucan 4B column was first equilibrated with Tris-HCl buffer (containing 1 M NaCl, 1.0 mM CaCl 2 , 1.0 mM MgCl 2 and 0.2 ml Tween-20) and 0.2 ml of serum was added. The above buffer eluted. The first 3.0 ml was discarded and 5.0 ml of components containing non-ConA bound AFP were collected.
