目的 ABCC8是ATP-敏感性K+(KATP)通道的调节亚基(又称SUR1),本研究拟对ABCC8基因15th内含子与16th外显子剪切处C1799854T(NCBI数据库编码:RS1799854)位点变异多态性,与军人18周有氧训练指标是否存在关联进行比对。方法对102名某驻地士兵进行18周耐力训练,训练负荷为5000 m长跑,每周进行3次。负荷强度:以心率为标准监控,该心率与士兵个体通气无氧阈对应。运用跑台逐级递增负荷运动方法,对新入伍士兵训练前后最大摄氧量(maximal oxygen uptake,VO2max)相关指标、跑节省化(running economy,RE)相关指标进行测定。采用聚合酶链反应-限制酶片段长度多型性(ploymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)技术方法测定ABCC8基因C1799854T位点变异多态性。结果三种基因型的分布频率分别为CC:14(0.14),CT:52(0.51),TT:36(0.35),符合Hardy-Weinberg平衡。按基因型分组后,CC基因型人群训练的VO2max变化率、RE心率变化率显著性高于TT基因型人群,比较差异具有统计学意义(P<0.05),测定的其他表型指标与基因型的差异性均无统计学意义(P>0.05)。结论 ABCC8基因C1799854T多态性与有氧耐力初始值指标无关联性;ABCC8基因C1799854T多态性可作为VO2max、RE心率有氧耐力训练敏感性的标记,CC型群体具有更好的易训性。
Objective ABCC8 is a regulatory subunit of ATP-sensitive K + (KATP) channel (also known as SUR1). In this study, we plan to investigate the relationship between the 15th intron of ABCC8 gene and the C1799854T (NCBI database code: RS1799854) Mutation polymorphism, and military 18-week aerobic training indicators whether there is correlation between the alignment. Methods A total of 102 resident soldiers were trained in endurance training for 18 weeks. The training load was 5000 m long run and three times a week. Load intensity: The heart rate as the standard monitoring, the heart rate and the individual soldiers breathe anaerobic threshold corresponding. By using treadmill step-by-step load-bearing method, the indexes of maximal oxygen uptake (VO2max) and running economy (RE) before and after training were obtained. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the polymorphism of C1799854T in ABCC8 gene. Results The frequency distribution of the three genotypes was CC: 14 (0.14), CT: 52 (0.51) and TT: 36 (0.35), respectively, which were in accordance with Hardy-Weinberg equilibrium. According to the genotypes, VO2max rate and RE rate of heart rate in CC genotype were significantly higher than those in TT genotype (P <0.05). The other phenotypic indexes and genotype The difference was not statistically significant (P> 0.05). Conclusion There is no association between C1799854T polymorphism of ABCC8 gene and the initial value of aerobic endurance. The C1799854T polymorphism of ABCC8 gene can be used as a marker of aerobic endurance training of VO2max and RE, and the CC genotype has better predictability.
