采用细胞单层培养方法,以cAMP为观察指标,研究肿瘤坏死因子(TNF-α)对正常和甲亢甲状腺细胞功能调控的特征。结果显示,(1)10~(-3)和10~(-1)U/ml的TNF-α对基础状态下正常甲状腺细胞生成cAMP的值无显著影响,当其浓度为10~3和10~3U/ml时,cAMP释放明显增加。TNF-α在10~(-3)~10U/ml的剂量范围内,对甲亢甲状腺细胞基础cAMP的生成无明显影响,但10~3U/ml的TNF-α则显著促进甲亢甲状腺细胞产生cAMP。(2)在10~(-3)~10U/ml的浓度区间,TNF-α显著抑制TSH对正常和甲亢甲状腺细胞生成cAMP的刺激效应,10~3U/ml的TNF-α仅抑制TSH对正常甲状腺细胞的刺激作用,对甲亢甲状腺细胞无明显影响。提示正常与甲亢甲状腺的功能均受TNF的调节,但两种组织对TNF的反应性不尽相同。
Using cell monolayer culture method, the cAMP as an indicator to study the tumor necrosis factor (TNF-α) on the normal and hyperthyroidism thyroid cell function regulation characteristics. The results showed that (1) 10 ~ (-3) and 10 ~ (-1) U / ml of TNF-α had no significant effect on cAMP production in normal thyroid cells under basal conditions. Only 10 ~ 3U / ml of TNF- Thyroid cells stimulate the role of hyperthyroidism thyroid cells had no significant effect. It is suggested that the function of thyroid in normal and hyperthyroidism is regulated by TNF, but the reactivity of TNF in different tissues is different.
