本文报告了人胚肺成纤维细胞(CCL137)中M受体的生化药理学特性.用放射配基结合实验证实了细胞中含有可饱和、高亲和力以及立体结构特异性的M受体.细胞膜上的M受体与腺苷酸环化酶呈负性偶联.用药理学和生物化学方法证实细胞中的M受体为M_2亚型.亲和标记后行电泳分析证实受体的分子量为72kDa.以上工作表明GCL137细胞中存在丰富的功能均一的M_2受体,其数量明显高于其它来源的人的培养细胞系,并同时含有糖皮质激素受体,是研究M_2受体及研究不同受体系统之间相互调控作用的良好细胞模型.
In this paper, the biochemical and pharmacological properties of M receptor in human embryonic lung fibroblasts (CCL137) were reported. Radioligand binding experiments demonstrated that the cells contain saturable, high affinity and stereospecific M receptors. Of the M receptor was negatively coupled to adenylate cyclase.M pharmacological and biochemical methods confirmed that the M receptor in the cell was an M_2 subtype.The electrophoretic analysis of the affinity tag followed by electrophoresis confirmed that the acceptor had a molecular mass of 72 kDa. The above work shows that GCL137 cells are rich in uniform function of M 2 receptors, the number was significantly higher than those from other human cultured cell lines, and also contains glucocorticoid receptor, is to study the M 2 receptor and the study of different receptor systems Between the regulation of the role of a good cell model.
