目的:研究微波辅助提取香椿叶中总黄酮的工艺条件和提取物清除DPPH自由基的能力。方法:在单因素试验的基础上,采用L16(45)正交试验法对香椿叶的微波提取工艺进行优选。以黄酮提取量为主参考指标,提取物以黄酮计的清除DPPH的IC50值为次参考指标,考察乙醇体积分数、固液比、微波提取时间、提取温度、提取次数对香椿叶总黄酮提取量的影响。结果:最佳工艺条件为:20g原料,以料液比1∶12加入70%的乙醇溶液,在80℃下每次微波提取20min,共提取4次。在最佳参数组合下,每克香椿叶可提取总黄酮25.7705mg,提取物以黄酮计的清除DPPH的IC50值为37.1073。结论:该工艺是微波法提取香椿叶总黄酮的最佳工艺。
OBJECTIVE: To study the technological conditions of microwave-assisted extraction of total flavonoids from Toona sinensis leaves and the ability of extracts to scavenge DPPH free radicals. Methods: Based on the single factor experiment, the microwave extraction technology of Toona sinensis leaves was optimized by L16 (45) orthogonal test. Taking the flavone extraction as the main reference index, the IC50 value of flavonoids-scavenging DPPH was used as the secondary reference index. The effects of ethanol volume fraction, solid-liquid ratio, microwave extraction time, extraction temperature and extraction times on the extraction of total flavonoids from Toona sinensis leaves Impact. Results: The optimum conditions were as follows: 20g raw material, 70% ethanol solution was added at the ratio of solid to liquid 1:12, and extracted by microwave at 80 ℃ for 20 min each time. Under optimal parameters, total flavonoids per gram of Toona sinensis can be extracted 25.7705mg, the IC50 value of flavonoids scavenging DPPH was 37.1073. Conclusion: This process is the best microwave extraction technology of total flavonoids from Toona sinensis leaves.
