目的:利用PCR Array技术观察健脾祛痰化瘀中药对脾虚痰浊动脉粥样硬化(AS)巴马小型猪主动脉AS信号通路相关基因表达变化的影响,探讨健脾祛痰化瘀方防治AS的可能机制。方法:将15只巴马小型猪随机分为正常组、模型组和治疗组,每组5只,正常组普通饲料喂养,模型与治疗组给予高热量高脂饲料喂养并进行冠脉球囊挤压伴拉伤及跑步措施进行干预至24周,制备巴马小型猪病症结合脾虚痰浊AS模型,治疗组除上述造模措施外给予健脾祛痰化瘀方灌胃24周。呼吸机麻醉后取材,提取各组主动脉总RNA,应用AS信号通路PCR芯片检测健脾祛痰化瘀方对脾虚痰浊AS巴马小型猪主动脉AS信号通路相关基因mRNA表达变化的影响。结果:PCR Array结果显示:与空白组相比,模型组显著上调的基因共13个,如ACE、BAX、SELE等基因,显著下调的基因共16个,如BCL2A1、LDLR、LPL等基因;与模型组相比,治疗组显著上调的基因共14个,如BCL2A1、LDLR、BCL2等,显著下调的基因共13个如BAX、PTGS1、SOD1等。结论:健脾祛痰化瘀方可能参与调控主动脉AS信号通路中脂质代谢与凋亡等相关基因表达从而起到防治脾虚痰浊AS巴马小型猪AS形成及稳点斑块的作用。
Objective: To observe the effects of Chinese herbal medicine for invigorating the spleen and phlegm resolving stasis on the expression of AS-related genes in the aorta of Alabama miniaturist pig with phlegm-visceral phlegm turbid phlegm atypia of spleen deficiency by PCR Array technique and to explore the preventive and therapeutic effects of Jianpi Qutan Huayu Recipe Possible mechanism of AS. Methods: Fifteen Bama miniature pigs were randomly divided into normal group, model group and treatment group, with 5 rats in each group. Normal group fed with normal diet, model and treatment group were given high calorie and high fat diet, Compression with injury and running measures intervention to 24 weeks to prepare parmesan miniature pig disease combined spleen deficiency phlegm turbid AS model, the treatment group in addition to the above modeling measures given Jianpi Qutan Huayu Fang gavage for 24 weeks. The ventilator was harvested and the total RNA was extracted from the aorta of each group. The effect of Jianpi Qutan Huayu Recipe (ASQP) on mRNA expression of AS signal pathway related genes in the aorta of AS rats was studied by AS signal pathway PCR. Results: The results of PCR Array showed that compared with the blank group, there were 13 genes that were significantly up-regulated in the model group (16 genes such as ACE, BAX and SELE), such as BCL2A1, LDLR and LPL Compared with the model group, there were 14 genes that were significantly up-regulated in the treatment group, such as BCL2A1, LDLR and BCL2, and 13 genes were significantly down-regulated, such as BAX, PTGS1 and SOD1. Conclusion: Jianpi Qutan Huayu Recipe may be involved in the regulation of the expression of lipid metabolism and apoptosis related genes in the AS signal pathway of the aorta and thus play an important role in prevention and treatment of AS formation and stable plaques in the spleen deficiency and phlegm turbidity AS Bama miniature pigs.
