目的研究异叶败酱提取物作用于人白血病K562细胞的分子机制。方法以异叶败酱提取物作用于K562细胞后提取细胞总蛋白,用双向电泳对蛋白进行分离,对异叶败酱提取物处理前后的K562细胞总蛋白双向电泳图谱进行差异分析,通过质谱技术鉴定差异明显的蛋白质。结果通过对比分析异叶败酱提取物处理前后的白血病K562细胞总蛋白双向电泳图谱,发现异叶败酱提取物处理组中有23个蛋白点发生了明显改变。经质谱和Mascot软件分析,成功鉴定了3个蛋白质,分别为热休克蛋白90β,甘油醛-3-磷酸脱氢酶,血红蛋白。结论通过蛋白质组学技术,发现了异叶败酱提取物处理前后白血病K562细胞差异明显的蛋白质点23个,这些差异蛋白质可能是异叶败酱提取物对白血病K562细胞产生增殖抑制、凋亡诱导和诱导分化作用的关键蛋白质。
Objective To study the molecular mechanism of the extract of Paeonia lactiflora on human leukemia K562 cells. Methods The total protein of K562 cells was extracted with the extract of Paeonia lactiflora and the protein was separated by two-dimensional electrophoresis. The two-dimensional electrophoresis patterns of total proteins of K562 cells before and after treatment were compared and analyzed by mass spectrometry Identification of significant differences in the protein. Results By comparing the two-dimensional gel electrophoresis of leukemia K562 cells before and after treatment with Paeonia lactiflora extract, 23 protein spots in the Paeonia lactiflora extract treatment group were significantly changed. By mass spectrometry and Mascot software analysis, successfully identified three proteins, namely heat shock protein 90β, glyceraldehyde-3-phosphate dehydrogenase, hemoglobin. Conclusion Through proteomic techniques, 23 different protein spots were found in K562 leukemia cells before and after treatment with the extract of Paeonia lactiflora. These differential proteins may be the result of the proliferation inhibition and apoptosis induction of Leukemia K562 cells And differentiation-induced differentiation of key proteins.
