目的 建立并鉴定脐血来源树突状细胞(DC) 培养体系, 在体外诱导脐血干细胞生成成熟DC,探讨其在抗肿瘤免疫中的作用。方法 脐血CD34 +来源的DC 以两步法, 即0 天开始使用GM CSF、TNF α、SCF、TPO、FL20, 8~14天用GM CSF、TNF α和IL 4诱导生成。并分别在形态(光学显微镜+电镜)、表面标记(流式细胞仪) 和功能(同种混合淋巴细胞反应) 上加以鉴定。结果 在合适的细胞因子组合下, 能够经过脐血干细胞培养得到成熟的DC, 具备典型的DC形态并在体外有效刺激同种淋巴细胞增殖。结论 通过合适的培养可以获得大量成熟DC。
Objective To establish and identify dendritic cells (DCs) culture system derived from umbilical cord blood and induce cord blood stem cells to generate mature DCs in vitro and to explore its role in anti-tumor immunity. Methods Cord blood CD34 + -derived DCs were induced by GM CSF, TNFα, SCF, TPO, FL20 starting from day 0 and GM CSF, TNFα and IL 4 from day 8 to day 14 in two steps. And identified in morphology (light microscope + electron microscope), surface marker (flow cytometry) and function (allomix lymphocyte reaction). Results Under suitable combinations of cytokines, mature DCs can be obtained by culturing umbilical cord blood stem cells, have typical DC morphology and effectively stimulate allogeneic lymphocyte proliferation in vitro. Conclusion A large number of mature DCs can be obtained by appropriate culture.
