目的研究资木瓜乙酸乙酯萃取物对不同抗原诱导的RBL-2H3细胞脱颗粒的影响,并初步探讨其作用机制。方法采用MTT法测定不同浓度资木瓜乙酸乙酯萃取物对细胞活性的影响,采用底物法检测不同浓度资木瓜乙酸乙酯萃取物对不同抗原引起的细胞内β-氨基己糖苷酶释放的影响,采用流式细胞分析仪检测不同浓度资木瓜乙酸乙酯萃取物对细胞表面受体FcεRIα表达的影响。结果在一定浓度范围内,对细胞活性没有显著性影响的资木瓜乙酸乙酯萃取物可显著抑制细胞内β-氨基己糖苷酶的释放和降低细胞表面受体FcεRIα的表达,且呈量效关系。结论资木瓜乙酸乙酯萃取物对RBL-2H3细胞脱颗粒具有显著的抑制作用,其作用可能与RBL-2H3细胞表面受体FcεRIα的表达降低有关。
Objective To study the effect of ethyl papaver euchroma pluvialis extract on the degranulation of RBL-2H3 cells induced by different antigens and to explore its mechanism. Methods MTT assay was used to determine the effect of different concentrations of ethyl acetate extract on the cell viability. Substrate method was used to detect the effect of different concentrations of papaya extract on the intracellular β-hexosaminidase released by different antigens Flow cytometry was used to detect the effect of different concentrations of papaver eupate on the expression of cell surface receptor FcεRIα. Results In a certain concentration range, papaya ethyl acetate extract, which had no significant effect on cell activity, significantly inhibited the release of β-hexosaminidase and decreased the expression of cell surface receptor FcεRIα in a dose-dependent manner . Conclusion The extract of papaya extract has a significant inhibitory effect on the degranulation of RBL-2H3 cells, which may be related to the decrease of the expression of FcεRIα on the surface of RBL-2H3 cells.
