目的:探讨E-cadherin在结直肠癌(colorectal carcinoma,CRC)中的表达及其与侵袭转移的关系。方法:采用免疫组织化学SABC法及原位杂交技术检测E-cadherin mRNA和蛋白在大肠正常黏膜、大肠腺瘤和CRC组织中的表达。结果:E-cadherin的mRNA和蛋白在CRC中的阳性率显著低于正常黏膜组织和腺瘤(P<0.01);随着CRC分化的降低和Dukes分期的增加,E-cadherin表达阳性率降低;E-cadherin的阳性率在有淋巴结和远处器官转移的CRC中均显著低于未转移组(P<0.01,P<0.05)。结论:E-cadherin表达减少或缺失在CRC的侵袭转移过程中发挥重要的作用。
Objective: To investigate the expression of E-cadherin in colorectal carcinoma (CRC) and its relationship with invasion and metastasis. Methods: The expression of E-cadherin mRNA and protein in colorectal mucosa, colorectal adenoma and CRC tissues were detected by immunohistochemical SABC method and in situ hybridization. Results: The positive rate of E-cadherin mRNA and protein in CRC was significantly lower than that in normal mucosa and adenoma (P <0.01). The positive rate of E-cadherin expression was decreased with the decrease of CRC and the increase of Dukes stage. The positive rate of E-cadherin in CRC with lymph node and distant organ metastasis was significantly lower than that in non-metastasis group (P <0.01, P <0.05). Conclusion: The decrease or deletion of E-cadherin plays an important role in the invasion and metastasis of CRC.
