作者用杂交瘤技术获得了纯系细胞株,能稳定分泌抗BMP单克隆抗体。该抗体能与人的骨和牙胚发生反应,具有很高的特异性。免疫组化染色结果表明,BMP存在于骨基质的胶原纤维、新生的骨基质、骨母细胞和骨髓中的未分化间充质细胞中;在人的牙胚中,BMP主要分布在前期牙木质、外釉细胞和造釉细胞以及形成牙槽骨的牙囊细胞中,表明骨的生长和形成以及牙齿的发育都与BMR密切相关。bBMP-McAb能与人的骨和牙胚发生反应,证实人和牛骨的BMP的抗原决定簇是相同的。BMP活性阻断实验证明,新研制的单克隆抗体能有效地封闭BMP活性基因。利用bBMP McAb这一特点,不仅可以纯化BMP,而且可以用来抑制骨的生长,为临床治疗骨肉瘤和其它骨疾病开拓了广阔的前景。
The authors obtained a line of pure lines using the hybridoma technique that secretes anti-BMP monoclonal antibodies stably. The antibody reacts with human bone and tooth germ and has high specificity. The results of immunohistochemistry showed that BMP was found in collagen fibers of bone matrix, newborn bone matrix, osteoblastic cells and undifferentiated mesenchymal cells in bone marrow. In human tooth germ, BMP mainly distributed in predental dental wood , Enamel and ameloblasts, and alveolar bone-forming dental follicles, indicating that bone growth and development and tooth development are closely related to BMR. bBMP-McAb reacts with human bone and tooth germ, confirming that the human and bovine BMP antigenic determinants are identical. BMP activity blocking experiments show that the newly developed monoclonal antibody can effectively block the BMP activity gene. The use of bBMP McAb not only can purify BMP, but also can inhibit bone growth, which opens up a broad prospect for the clinical treatment of osteosarcoma and other bone diseases.
