硝酸甘油(GTN)降低心脏前负荷和动脉阻力,因此,舌下给 GTN 治疗急性冠心病发作和左心室衰竭所致的肺水肿是非常有效的。用药后血液动力学作用一般在1~2min即可出现,5~10min 达最大。继 GTN 胶囊后,喷雾剂也已作为舌下给药的一种方式。以往的喷雾剂含对身体有明显伤害作用的氟氯烃(FCH),最近一种无 FCH 而通过泵系统释放GTN 的制剂已问世。为证明它与含 FCH 制剂具有相等的生物效价,研究了应用两种喷雾剂的 GTN 生物利用度。16名健康受试者舌下给不同 GTN 喷雾齐0 2×0.41mg(A 剂含 FCH,B 剂无FCH),测定 GTN 生物等效量。2~30min内多次取血测定 GTN 的血浆浓度(GC 法)
Nitroglycerin (GTN) reduces preload and arterial resistance, therefore sublingual GTN is very effective in treating pulmonary edema caused by episodes of acute coronary artery disease and left ventricular failure. After treatment, hemodynamic effects generally appear in 1 ~ 2min, 5 ~ 10min up to maximum. Following the introduction of GTN capsules, sprays have also been used as a means of sublingual administration. Previous sprays contained hydrochlorofluorocarbons (FCHs), which have a significant body-damaging effect, and more recently a formulation that releases GTN through a pump system without FCH has been available. To demonstrate that it has equivalent biological potency to FCH-containing formulations, the GTN bioavailability of two sprays was studied. Twenty-six healthy subjects underwent sublingual gonadotropin spray with different GTN doses of 0 2 × 0.41 mg (FCH for group A and no FCH for group B) to determine the bioequivalence of GTN. 2 ~ 30min multiple blood samples were taken to determine the plasma concentration of GTN (GC method)
