目的:观察葛根素对慢性脑缺血大鼠海马CA1区NGF表达的影响。方法:将84只体重在200~300 g的健康雄性大鼠,其中随机挑选20只,分成2个亚组,分别为30 d(1月组)空白组和90 d(3月组)空白组(正常对照),其余64只大鼠使用永久性双侧颈总动脉结扎法制作慢性脑缺血模型并随机又分成3个组,分别为模型组、雌二醇组和葛根素治疗组,每组又分别分成30 d(1月组)和90 d(3月组)2亚组。雌二醇组使用雌二醇ip治疗大鼠慢性脑缺血(1 mg.kg-1.d-1);葛根素治疗组使用葛根素灌服治疗大鼠(50 mg.kg-1.d-1);相同剂量的生理盐水用于模型组大鼠。各组分别在30,90 d后处死。用免疫组化方法观察大鼠海马CA1区神经生长因子(NGF)的表达,应用显微图像分析系统进行分析。结果:30 d模型组,雌二醇组、葛根素组与正常组比较,海马CAl区可见NGF的表达明显升高(P<0.05);雌二醇组、葛根素组与模型组比NGF的表达明显升高(P<0.05)。90 d(3月组)模型组、雌二醇、葛根素组大鼠海马CAl区NGF的表达与正常组比较NGF的表达上调(P<0.05),与90 d模型组比较,雌二醇大鼠海马CAl区NGF的表达无显著性差异,葛根素组大鼠海马CAl区的NGF表达明显上调(P<0.05)。结论:葛根素可能通过上调NGF水平,发挥对脑缺血神经元的保护作用。
Objective: To observe the effect of puerarin on NGF expression in hippocampal CA1 area of rats with chronic cerebral ischemia. Methods: A total of 84 healthy male rats weighing 200-300 g were randomly divided into two subgroups: 30 days (January group) and 90 days (March group) (Normal control). The remaining 64 rats were randomly divided into 3 groups: model group, estradiol group and puerarin-treated group Groups were divided into 30 d (January group) and 90 d (March group) 2 subgroups. In the estradiol group, estradiol ip was used to treat rats with chronic cerebral ischemia (1 mg.kg-1.d-1). Puerarin-treated rats were treated with puerarin (50 mg.kg-1.d -1). The same dose of saline was used in model rats. Each group was sacrificed 30 and 90 days later. Immunohistochemistry was used to observe the expression of nerve growth factor (NGF) in rat hippocampal CA1 region, and then analyzed by microscopic image analysis system. Results: Compared with the normal group, the expression of NGF in hippocampal CA1 region was significantly increased in the 30 d model group, estradiol group and puerarin group (P <0.05). Compared with the NGF group The expression was significantly increased (P <0.05). Compared with the normal group, the expression of NGF in hippocampal CA1 region of model group, estradiol and puerarin group at 90 d (March group) was up-regulated (P <0.05). Compared with 90-day model group, There was no significant difference in the expression of NGF in hippocampal CA1 region of rats, while the expression of NGF in hippocampal CA1 region of rats in puerarin group was significantly increased (P <0.05). Conclusion: Puerarin may play a neuroprotective role in cerebral ischemia by up-regulating the level of NGF.
