表面污染和材料褐变是龙眼组织离体培养成活率低的主要原因,采用切取中间茎段内部组织培养和多种药液交替浸泡的前处理方法,是降低污染率、褐化率,成功建立无菌初代培养物的有效措施;不同物候期的茎段,诱芽率不同,以夏、秋季的茎段较为适合作为起始外植体;选用MS基本培养基诱芽,诱芽率可达73%左右,且生长整齐,叶色较绿,为以脱毒为目的的龙眼茎尖培养研究提供无菌、无褐变的茎尖。
