目的探讨应用重组人粒细胞集落刺激因子(rHuG-CSF)动员外周血干细胞时,异体、自体造血干细胞移植供、受者的骨髓及外周血CD3+4细胞和T细胞亚群的动态变化特点及最佳外周血干细胞采集时间。方法对兰州军区兰州总医院2001~2002年间的12名健康供者和16例化疗7d后的恶性血液病患者,以rHuG-CSF每12h150μg,皮下注射,连用5d。应用流式细胞仪测定骨髓及外周血CD3+4细胞和T细胞亚群。结果(1)应用rHuG-CSF前,两组的骨髓CD3+4细胞差异有显著性意义(P<0·01)。应用48h以后,两者的骨髓CD3+4细胞均较前有明显增加(P<0·01),两组之间差异仍存在显著性意义(P<0·01)。两者的骨髓CD3+4细胞高峰值出现在96h后,较应用前差异有显著性意义(P<0·01)。(2)应用rHuG-CSF72h以后,两组外周血中CD3+4细胞均较前有明显增加(P<0·01)。96h以后,两组的CD3+4细胞达高峰,较应用前差异有显著性意义(P<0·01)。(3)应用rHuG-CSF对两者的T淋巴细胞亚群无明显影响。结论应用rHuG-CSF动员外周血干细胞96h以后,异体、自体造血干细胞移植供、受者的骨髓及外周血CD3+4细胞均达到高峰,可适时采集外周血干细胞。
Objective To investigate the dynamic changes of CD3 + 4 cells and T lymphocyte subsets in bone marrow and peripheral blood of allogeneic and autologous stem cell transplantation recipients mobilized peripheral blood stem cells using recombinant human granulocyte colony-stimulating factor (rHuG-CSF) Best peripheral blood stem cell collection time. Methods Twelve healthy donors from Lanzhou General Hospital of Lanzhou Military Region from 2001 to 2002 and 16 patients with hematologic malignancies after chemotherapy for 7 days were injected subcutaneously with rHuG-CSF every 12 hours for 5 days. Flow cytometry was used to detect CD3 + 4 cells and T lymphocyte subsets in bone marrow and peripheral blood. Results (1) Before using rHuG-CSF, the difference of CD3 + 4 cells in bone marrow between the two groups was significant (P <0.01). After 48 hours of application, the CD3 + 4 cells in both groups were significantly increased compared with those before (P <0.01). There was still significant difference between the two groups (P <0.01). The peak of CD3 + 4 cells in both groups appeared after 96 h, which was significantly different from that before application (P <0.01). (2) After rHuG-CSF was administered for 72 hours, CD3 + 4 cells in peripheral blood of both groups increased significantly (P <0.01). After 96h, the CD3 + 4 cells peaked in both groups, which was significantly different from that before application (P <0.01). (3) The application of rHuG-CSF had no significant effect on the T lymphocyte subsets. Conclusion After 96 h mobilization of peripheral blood stem cells by rHuG-CSF, CD3 + 4 cells in bone marrow and peripheral blood of allogeneic and autologous stem cell transplantation recipients reached their peak, and peripheral blood stem cells can be collected in a timely manner.
