目的探讨二仙汤、二至丸和地黄煎对小鼠脾细胞和卵巢颗粒细胞的调节作用。方法取小鼠脾脏和卵巢分别制备脾单个核细胞和卵巢颗粒细胞,于24孔板中培养,分别加入二仙汤、二至丸以及地黄煎的水提物和醇提物,以香菇多糖水溶剂为阳性对照药物,运用MTT法观察细胞增殖情况;酶联免疫吸附法(ELISA)测定细胞上清液中IL-1、IFN-γ及E2的浓度。结果二仙汤、二至丸、地黄煎各组对脾单个核细胞及卵巢颗粒细胞均具有较好的增殖作用(P<0.05,P<0.01);三方均可提高小鼠脾单个核细胞培养上清液中IL-1、IFN-γ的含量(P<0.05,P<0.01),但与阳性对照组相比,三方均无差异(P>0.05);三方均可提高小鼠卵巢颗粒细胞上清液中的E2水平(P<0.01,P<0.05);与阳性对照组相比,三方均无差异(P>0.05)。结论二仙汤、二至丸、地黄煎对小鼠脾细胞和卵巢颗粒细胞均有较好的调节作用。
Objective To investigate the regulation effect of Erxian Decoction, Erzhi Pills and Dihuang Decoction on mouse spleen cells and ovarian granulosa cells. Methods Splenic mononuclear cells and ovarian granulosa cells were prepared from the spleen and ovary of mice and cultured in 24-well plates. The water extract and ethanol extract of Erxian decoction, Erzhi decoction, and Radix Rehmanniae Decoction were respectively added, Solvents were positive control drugs, MTT assay was used to observe the cell proliferation. ELISA was used to determine the concentration of IL-1, IFN-γ and E2 in the cell supernatant. Results Both Er-xian decoction, Erzhi pill and Rehmannia glutinosa decoction had better proliferation activity on spleen mononuclear cells and ovarian granulosa cells (P <0.05, P <0.01). All three groups could improve the culture of mouse spleen mononuclear cells (P <0.05, P <0.01), but there was no significant difference among the three groups (P> 0.05). All three groups could increase the level of IL-1 and IFN- The level of E2 in the supernatant (P <0.01, P <0.05) showed no significant difference among the three groups (P> 0.05). Conclusion Erxian decoction, Erzhi pill and Rehmannia glutinosa decoction have better regulation on mouse spleen cells and ovarian granulosa cells.
