目的:研究给予外源性表皮生长因子(EGF)对完全胃肠外营养(TPN)大鼠小肠隐窝EGF受体表达的影响以及与小肠粘膜细胞增殖的关系.方法:采用大鼠TPN模型,小肠组织切片HE染色,测定隐窝细胞数及有丝分裂指数;采用免疫组织化学结合计算机图像分析的方法,测定隐窝细胞EGF含量及EGF受体的表达.实验共使用200~250g体重SD大鼠24只,随机分组:对照组(n=8),仅行虚拟手术,即行右颈静脉结扎,每日饲以普通饲料及水;TPN组(n=8),禁食水,输注常规TPN;TPN-EGF组(n=8),输注常规TPN溶液,并每日按0.1μg/(次·g)皮下注射EGF2次.各组均观察14d.结果:TPN大鼠小肠隐窝EGF含量及EGF受体表达显著减少(P<0.01),隐窝细胞数和有丝分裂指数也明显降低(P<0.05);当给予外源性EGF后,隐窝EGF含量、EGF受体表达以及隐窝细胞数、有丝分裂指数均未降低.结论:EGF能预防TPN大鼠小肠隐窝EGF受体表达及EGF含量下降,维持细胞生成率.
OBJECTIVE: To study the effect of exogenous epidermal growth factor (EGF) on the expression of EGF receptor in the small intestine crypt of complete parenteral nutrition (TPN) rats and its relationship with the proliferation of intestinal mucosal cells. METHODS: Rat TPN model was used to detect the number of crypt cells and mitotic index by HE staining of small intestine tissue sections. EGF content and EGF receptor expression in crypt cells were measured by immunohistochemistry and computer image analysis. A total of 24 SD rats weighing 200-250 g were randomly divided into control group (n=8). Only virtual surgery was performed, ie, right jugular vein ligation was performed. Normal feed and water were fed daily; TPN group (n=8). ), Fasting water, infusion of conventional TPN; TPN-EGF group (n = 8), infusion of conventional TPN solution, and daily by 0.1μg / (times g) subcutaneous injection of EGF 2 times. All groups were observed 14d. RESULTS: The EGF content and EGF receptor expression in small intestine crypts of TPN rats were significantly decreased (P<0.01), and the number of crypt cells and mitotic index were also significantly decreased (P<0.05); after administration of exogenous EGF The crypt EGF content, EGF receptor expression, crypt cell number and mitotic index were not decreased. Conclusion: EGF can prevent the expression of EGF receptor and the content of EGF in the intestinal crypt of TPN rats, and maintain the cell formation rate.
