目的:了解雷公藤甲素诱导大鼠肾上腺皮质细胞凋亡的发生。方法:体外分离培养大鼠肾上腺皮质细胞,以蛋白因子添加素V和碘化丙啶(PI)联合标记,流式细胞仪检测,观察雷公藤甲素诱导细胞凋亡的特征。结果:雷公藤甲素以时间和剂量依赖的方式诱导细胞凋亡,0、12.5、25、50、100和200μg/L雷公藤甲素处理细胞2h,凋亡率分别为1.67%、16.54%、32.53%、47.62%、57.12%及77.45%;100μg/L雷公藤甲素染毒0、0.5、1、2、3和4h,细胞凋亡率分别为1.12%、14.37%、35.26%、49.75%、62.65%及78.46%。结论:雷公藤甲素可诱导肾上腺皮质细胞凋亡。
Objective: To understand the triptolide-induced apoptosis of adrenocortical cells in rats. Methods: Adrenal cortical cells were isolated and cultured in vitro. Combined with the addition of protein V and propidium iodide (PI), the characteristics of apoptosis induced by triptolide were observed by flow cytometry. Results: Triptolide induced apoptosis in a time-and dose-dependent manner. The apoptotic rates of triptolide treated with 0, 12.5, 25, 50, 100 and 200 μg / L triptolide were 1.67% and 16.54% 32.53%, 47.62%, 57.12% and 77.45% respectively. The apoptotic rates of triptolide treated with 100 μg / L triptolide were 1.12%, 14.37%, 35.26% and 49.75% at 0, , 62.65% and 78.46% respectively. Conclusion: Triptolide can induce apoptosis of adrenocortical cells.
