目的比较人表浅性与浸润性膀胱癌基因表达谱的差异。方法应用基因芯片技术对表浅性膀胱癌和浸润性膀胱癌的 RNA 进行检测分析。得到芯片灰度扫描图,通过图像处理软件GenePix Pro 3.0对图像进行分析,得到表达差异基因的相应数据,根据差异基因的类型及膀胱癌的特点分析数据。结果按显著差异基因标准,从13939条基因中筛选出显著差异表达基因:表浅组714条,其中480条下调,234条上调;浸润组470条,其中302条下调,168条上调。在显著差异基因中,下调基因的数量比上调基因多近1倍,免疫相关基因在浸润性膀胱癌中下调最明显。结论浸润性和表浅性膀胱癌的基因表达谱具有许多共同特点,但表浅性膀胱癌的差异表达基因较浸润性膀胱癌更多。免疫相关基因在浸润性膀胱癌中缺失较表浅性膀胱癌严重。
Objective To compare the gene expression profiles between superficial and invasive bladder cancer. Methods The RNA of superficial bladder cancer and invasive bladder cancer were detected by gene chip technique. The gray scale scan of the chip was obtained. The image was analyzed by image processing software GenePix Pro 3.0 to get the corresponding data of the differentially expressed genes. The data were analyzed according to the types of the differentiated genes and the characteristics of bladder cancer. Results According to the criteria of significant difference, significant differentially expressed genes were screened out from 13,939 genes: 714 in the superficial group, of which 480 were down-regulated and 234 were up-regulated. There were 470 infiltration groups, of which 302 were down-regulated and 168 were up-regulated. Among the significantly different genes, the number of down-regulated genes is nearly double that of up-regulated genes, and the immune-related genes are most obviously down-regulated in invasive bladder cancer. Conclusion The gene expression profiles of invasive and superficial bladder cancer have many common features, but there are more differentially expressed genes in superficial bladder cancer than invasive bladder cancer. Immune-related genes in invasive bladder cancer is more severe than superficial bladder cancer.
