目的:采用HPLC分析测定28个不同采集地土茯苓(断面红色、白色)中落新妇苷的含量,并通过对土茯苓(断面红色、白色)渗透型炎症的研究,拟对该品种进行药效与成分的评价。方法:RP-HPLC测定落新妇苷含量,采用依利特Hy-persil ODS2色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.1%冰醋酸溶液(21∶79),流速1.0 mL.min-1,检测波长291 nm,柱温25℃,理论塔板数以落新妇苷计不得低于5 000。用毛细血管通透性试验比较不同断面颜色药的抗炎作用。结果:在选定的色谱条件下,落新妇苷在0.003 379~4.004μg呈良好线性关系。加样回收实验加样80%,100%,120%,平均回收率分别为100.1%,101.9%,99.3%。断面白色土茯苓中落新妇苷质量分数为0.19%~2.46%,断面红色土茯苓中落新妇苷质量分数为2.10%~5.92%。药效学实验结果表明,断面白色土茯苓抗炎抑制率为32%,断面红色土茯苓抗炎抑制率仅为21%。结论:红、白2种断面土茯苓中落新妇苷的含量及抗炎作用均存在较大差异,土茯苓中落新妇苷的含量高低与其抗炎作用的强弱不呈正相关。
OBJECTIVE: To determine the content of astilbin in Poria cocos (the red and white section) of 28 different provenances by HPLC and to study its efficacy on the infiltration inflammation of Poria cocos (cross-section red and white) And the composition of the evaluation. Methods: The content of astilbin was determined by RP-HPLC. The mobile phase was acetonitrile-0.1% glacial acetic acid (21:79), the flow rate was 1.0 mL · min using Hy- persil ODS2 column (4.6 mm × 250 mm, -1, the detection wavelength is 291 nm, the column temperature is 25 ℃, and the number of theoretical plates should not be less than 5 000 in terms of astilbin. Results: Under the selected chromatographic conditions, astilbin showed a good linearity at 0.003 379 ~ 4.004μg. Replenishment experiments with sample loading 80%, 100%, 120%, the average recovery rates were 100.1%, 101.9%, 99.3%. The content of astilbin in the section of white tuckahoe was 0.19% -2.46%, and the content of astilbin in the section of red tuckahoe was 2.10% -5.92%. The results of pharmacodynamic experiments showed that the anti-inflammatory inhibitory rate of Poria Cotosum was 32% and the anti-inflammatory inhibition rate of Poria cochinchinensis was only 21%. CONCLUSION: The content of astilbin and the anti-inflammatory effect of Smilax china both in red and white sections are quite different. The content of astilbin in Smilax china is not positively correlated with the anti-inflammatory effect.
