非小细胞肺癌组织中HIF-1α和BNIP3的表达及相关性研究

目的:研究缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)和Bcl-2/腺病毒E1B19kDa结合蛋白3(Bcl-2andadenovirus E1B19kDa interacting protein3,BNIP3)在非小细胞肺癌(non-small cell lung carcinoma,NSCLC)组织中的表达及其相关性。方法:收集45例新鲜NSCLC标本(含癌旁组织)和14例正常对照肺组织,采用半定量RT-PCR和SP免疫组织化学方法,测定HIF-1α和BNIP3的表达水平及分析二者相关性,并与组织类型、分化程度、淋巴结转移、TNM分期、年龄、吸烟等进行多因素分析。结果:肺癌肿瘤组织中HIF-1α、BNIP3的mRNA表达水平显著高于癌旁组织及正常对照肺组织(P<0.01),并且两者表达呈正相关(r=0.68,P<0.01);HIF-1α、BNIP3在肿瘤组织中的蛋白阳性表达率分别为53.3%、66.7%,显著高于癌旁组织及正常对照肺组织(P<0.01),且二者阳性率呈正相关(rs=0.38,P<0.01)。单因素分析发现HIF-1α与组织类型、TNM分期相关(P<0.05),而BNIP3在相关临床病理参数分组中无显著差异。结论:HIF-1α与BNIP3在NSCLC中高表达且呈正相关,推测二者的高表达在肺癌的发生发展中可能具有重要作用。 Objective: To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and Bcl-2 / adenovirus E1B19kDa binding protein 3 (Bcl-2andadenovirus E1B19kDa interacting protein3) in non-small cell lung cancer cell lung carcinoma (NSCLC)) and its correlation. Methods: Forty-five fresh NSCLC specimens (including paracancerous tissues) and 14 normal control lung tissues were collected. The expression of HIF-1α and BNIP3 were detected by semi-quantitative RT-PCR and SP immunohistochemistry. , And multivariate analysis was performed with the type of tissue, degree of differentiation, lymph node metastasis, TNM stage, age, smoking and so on. Results: The mRNA expression levels of HIF-1α and BNIP3 in lung cancer tissues were significantly higher than those in para-cancerous tissues and normal lung tissues (P <0.01), and there was a positive correlation between them (r = 0.68, P <0.01) 1α and BNIP3 were 53.3% and 66.7% respectively, which were significantly higher than those in para-cancerous tissue and normal lung tissue (P <0.01), and the positive rates of both were positively correlated (rs = 0.38, P <0.01). Univariate analysis revealed that HIF-1alpha correlated with histological type and TNM stage (P <0.05), while there was no significant difference between BNIP3 and clinical pathological parameters. Conclusion: HIF-1α and BNIP3 are highly expressed in NSCLC and positively correlated with each other. It is speculated that the high expression of both may play an important role in the development of lung cancer.
