目的:观察赤雹根总皂苷对II型胶原性关节炎(CIA)模型大鼠的治疗作用及对CD4~+、CD8~+ T细胞表达的影响。方法:雄性Wistar大鼠80只,随机取10只作为正常对照组,其余大鼠尾根部多点皮内注射牛II型胶原和完全弗氏佐剂的乳化剂,7天后加强免疫一次,建立CIA大鼠模型,以关节炎指数(AI)≧6为建模成功。将模型大鼠按AI值水平随机分为赤雹根总皂苷80、40、20mg/kg组、雷公藤多苷12mg/kg组和模型对照组,每组11只。各给药组连续ig给药35天,模型对照组合正常对照组ig等体积的蒸馏水。分别于给药前及给药后7、14、21、28、35d测定大鼠四肢踝关节肿胀度和计算关节炎指数。末次给药后,取血清、脾脏,计算脾脏指数(SI);ELISA及Western Blot法分别检测CIA大鼠血清及脾脏中T淋巴细胞亚群CD4~+、CD8~+ T细胞的蛋白表达水平。结果:给药第35d,赤雹根总皂苷80、40、20mg/kg组踝关节肿胀度、AI值、SI值均显著低于模型对照组;血清及脾脏中CD4~+T细胞的蛋白表达水平、CD4~+/CD8~+的比值均显著低于模型对照组;CD8~+ T细胞的蛋白表达水平均显著高于模型对照组。结论:赤雹根总皂苷对RA有较好的治疗作用,调节CD4~+、CD8~+ T淋巴细胞的水平是其治疗RA的主要机制之一。
Objective: To observe the therapeutic effect of total alkaloids from red hail root on type Ⅱ collagen-induced arthritis (CIA) rats and its effect on the expression of CD4 ~ +, CD8 ~ + T cells. Methods: Eighty male Wistar rats were randomly selected as the normal control group. The remaining rats were intradermally injected with bovine type II collagen and complete Freund’s adjuvant emulsified agent, Rat model with arthritis index (AI) ≧ 6 was modeled successfully. The model rats were randomly divided into AI groups according to the AI value of 80, 40, 20 mg / kg groups, 12 mg / kg of tripterygium glycosides and model control group, 11 rats in each group. Each administration group was administered ig for 35 days continuously, and the model control group was given the same volume of distilled water as the normal control group. The degree of ankle swelling in all limbs and the arthritis index were measured before administration and at 7, 14, 21, 28 and 35 d after administration. After the last administration, serum and spleen were collected to calculate the spleen index (SI). The levels of CD4 ~ + and CD8 ~ + T cells in serum and spleen of CIA rats were detected by ELISA and Western Blot respectively. Results: On the 35th day after administration, the degree of ankle swelling, AI and SI of 80, 40 and 20 mg / kg groups were significantly lower than those of model control group. The protein expression of CD4 ~ + T cells in serum and spleen The level of CD4 ~ + / CD8 ~ + was significantly lower than that of the model control group. The protein expression level of CD8 ~ + T cells was significantly higher than that of the model control group. Conclusion: The total saponins of red hail gum have a good therapeutic effect on RA, and the regulation of the levels of CD4 ~ + and CD8 ~ + T lymphocytes is one of its main mechanisms in the treatment of RA.
