目的探讨麝香促进骨缺损愈合的可能作用机制。方法 300只SD大鼠以颅骨钻制备颅骨骨缺损模型,造模后随机分为模型组和实验组,每组150只。两组再按照给药时间分为第7、14、28天3个时间点,每个时间点50只。实验组大鼠每天灌服天然麝香4.2 mg/100 g,模型组每天灌服等量的生理盐水,均每日灌胃1次。分别于灌胃第7、14、28天采用实时荧光定量PCR法检测两组大鼠骨缺损处组织中转化生长因子β(TGF-β)和血管内皮生长因子(VEGF)mRNA水平。结果与模型组同时间点比较,实验组第7、28天TGF-βmRNA及第28天VEGF mRNA表达显著升高(P<0.05);实验组第28天TGF-β、VEGF mRNA表达明显高于本组第14天(P<0.05)。结论麝香可有效促使大鼠颅骨骨缺损区的愈合,其机制可能与升高缺损处骨组织TGF-β、VEGF mRNA表达有关。
Objective To investigate the possible mechanism of Musk promoting bone defect healing. Methods 300 SD rats were randomly divided into model group and experimental group with 150 skulls. The two groups were divided into three time points on the 7th, 14th and 28th days according to the administration time, and each time point was 50. The rats in the experimental group were given natural musk 4.2 mg / 100 g daily. The rats in the model group were orally administered with the same amount of normal saline daily. The levels of TGF-β and VEGF mRNA in bone defects of the two groups were detected by real-time fluorescence quantitative PCR at the 7th, 14th and 28th day respectively. Results Compared with the model group, the mRNA expression of TGF-β and VEGF at the 28th and 28th day in the experimental group were significantly increased (P <0.05). The expression of TGF-β and VEGF mRNA in the experimental group at 28th day was significantly higher than that of the experimental group The group 14 days (P <0.05). Conclusion Musk can effectively promote the healing of the skull defects in rats. The mechanism may be related to the increase of the expression of TGF-β and VEGF mRNA.
