目的:建立高效液相色谱法测定刺玫果中金丝桃苷的含量,为刺玫果的进一步开发和利用提供科学依据。方法:采用Dionex-C18柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸溶液(15∶85)为流动相,流速1.0 mL/min,柱温30℃,检测波长360 nm。结果:金丝桃苷在0.008~0.160 mg/mL浓度范围内与峰面积有良好的线性关系(r=0.9999),平均加样回收率为97.9%,RSD小于2.0%。结论:本文所建立的方法简便、快速、准确、重复性好,可用于刺玫果中金丝桃苷的含量测定。
OBJECTIVE: To establish a HPLC method for the determination of hyperoside in Prickly Pear, providing a scientific basis for the further development and utilization of Prickly Pear. METHODS: The mobile phase was acetonitrile-0.1% phosphoric acid solution (15:85) at a flow rate of 1.0 mL / min on a Dionex-C18 column (250 mm × 4.6 mm, 5 μm) with a detection wavelength of 360 nm at 30 ℃. Results: Hyperoside had a good linear relationship with the peak area (r = 0.9999) in the range of 0.008-0.160 mg / mL with an average recovery of 97.9% and a RSD of less than 2.0%. Conclusion: The method established in this paper is simple, rapid, accurate and reproducible. It can be used for the determination of hyperoside in Prickly pear fruit.
