研究‘库尔勒香梨’等19个主要梨品种的cp DNA trn S-trnf M区域的系统发育。利用trn S-trnf M区域的通用引物对总DNA进行扩增,将扩增出的PCR产物进行克隆测序,获得了长度为1 272~1 642 bp的19条序列。序列分析结果表明,‘库尔勒香梨’与白梨系统的同源性为85.01%,与新疆梨系统的同源性为78.60%,与西洋梨系统的同源性为78.28%,与砂梨系统的同源性为77.47%,与秋子梨系统的同源性为77.91%;基于测定序列构建的系统进化树表明,19个梨梨品种分为两大组,一组是‘库尔勒香梨’‘黑酸梨’‘句句梨’‘金川雪梨’、杜梨、‘京白’‘身不知’‘伏茄’‘冬巴’‘慈梨’;另一组是‘鸭梨’‘苹果梨’‘锦丰’‘砀山酥’梨、‘新世纪’‘南果梨’‘早酥’‘翠伏’和‘象牙’。
To study the phylogeny of cp DNA trn S-trnf M region in 19 major pear cultivars such as ’Korla Fragrans’. The total DNA was amplified by universal primers of trn S-trnf M region. The amplified PCR products were cloned and sequenced, and 19 sequences with length of 1 272-1 642 bp were obtained. The results of sequence analysis showed that the homology between ’Korla’ pear and Pear was 85.01%, that of Xinjiang pear system was 78.60%, that of ’Pear pear’ system was 78.28% The homology was 77.41% and 77.91% respectively. The phylogenetic tree based on the sequences indicated that 19 pear cultivars were divided into two groups, one was ’Korla fragrant pear’ Pear "’Jinchuan Sydney’, Pear, ’Jingbai’ body I do not know ’’ Fuwa ’’ Dongba ’’ Ciyi ’; the other group is’ pear ’apple pear’ ’Jinfeng’ ’Dangshan crispy’ pear, ’New Century’ ’South fruit pear’ ’early crisp’ ’Cuvu’ and ’ivory’.
