目的观察糖脂平对高脂饮食诱导的胰岛素抵抗(IR)大鼠血脂及脂肪因子的影响。方法将80只雄性SD大鼠随机分为正常组(20只)和造模组(60只),造模组用高脂高糖饲料喂养8周造模,其中54只造模成功的大鼠分为模型组、中药组、西药组,每组18只。中药组和西药组分别给予糖脂平和二甲双胍灌胃,模型组和正常组灌服等量的生理盐水。给药8周后,用ELISA测定各组大鼠的血清脂联素、瘦素、抵抗素水平,同时检测血脂和游离脂肪酸(FFA)水平。结果与正常组相比,模型组大鼠肥胖明显,总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、FFA均明显增高,高密度脂蛋白胆固醇(HDL-C)显著降低,表明模型组大鼠发生了脂代谢紊乱。同时脂肪因子、瘦素和抵抗素水平显著升高,脂联素水平显著降低。与模型组比较,糖脂平可以降低TG、LDL-C和FFA、瘦素、抵抗素水平,提高脂联素水平,对TC和HDL-C无明显影响。结论糖脂平可改善IR大鼠血脂紊乱,调节脂肪因子水平。
Objective To observe the effects of “Tang Zhi Ping” on blood lipids and adipokines in insulin resistance (IR) rats induced by high fat diet. Methods Eighty male Sprague-Dawley rats were randomly divided into normal group (n = 20) and model group (n = 60). The model group was fed with high-fat and high-sugar diet for 8 weeks, of which 54 rats For the model group, traditional Chinese medicine group, western medicine group, each group of 18. The traditional Chinese medicine group and the western medicine group were respectively administered with glycolipid and metformin, and the model group and the normal group were given the same amount of normal saline. After 8 weeks of administration, the levels of serum adiponectin, leptin and resistin in each group were measured by ELISA, and the levels of serum lipids and free fatty acids (FFA) were also measured. Results Compared with the normal group, the obese rats in the model group were significantly obese and the levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and FFA were significantly increased, while the levels of high density lipoprotein cholesterol -C) was significantly lower, indicating that lipid metabolism disorders occurred in the model group rats. At the same time, adiponectin, leptin and resistin levels were significantly increased, adiponectin levels were significantly lower. Compared with the model group, glycolipin can reduce TG, LDL-C and FFA, leptin, resistin levels, increase adiponectin levels, TC and HDL-C had no significant effect. Conclusion TGP can improve dyslipidemia and regulate the level of adipokines in IR rats.
