目的探讨金属基质蛋白酶-2(matrix metalloproteinase-2,MMP-2)基因启动子区-735C/T位点的基因多态性与病毒性脑膜炎易感性及血脑屏障(blood-brainbarrier,BBB)通透性关系。方法本研究应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法,测定了186例病毒性脑膜炎患者和202例健康者的MMP-2基因启动子区-735C/T位点的基因多态性,对病毒性脑膜炎组和对照组的基因频率以及不同基因型患者脑脊液白蛋白指数(CALB指数)进行对比分析。结果-735C/T位点基因型频率(CC、CT、TT)在病例组中分别为63.44%、31.72%、4.84%,在对照组分别为49.01%、42.57%、8.42%;病例组的C等位基因频率为79.30%、T等位基因频率为20.70%,对照组则为70.30%、29.70%,两者之间比较差异有统计学意义。而CALB指数在CC组(19.019±6.932),CT+TT组(15.835±6.047),两者比较差异有统计学意义(P=0.002)。结论 MMP-2基因启动子区-735C/T位点多态性可能与病毒性脑膜炎的易感性有关,其中C基因可能是易感基因,而T基因可能是保护基因。且该位点多态性可能通过影响BBB的通透性来影响病毒性脑膜炎的易感性。
Objective To investigate the association between the polymorphism of -735C / T in the promoter region of matrix metalloproteinase-2 (MMP-2) gene and the susceptibility to viral meningitis and the blood-brain barrier (BBB) Permeability relationship. Methods In this study, polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to detect the promoter region of -735C / T of MMP-2 gene in 186 patients with viral meningitis and 202 healthy controls Point gene polymorphism, viral meningitis group and control group gene frequency and different genotypes of patients with cerebrospinal fluid albumin index (CALB index) were compared. Results The frequency of genotypes (CC, CT, TT) at -735C / T was 63.44%, 31.72% and 4.84% in the case group and 49.01%, 42.57% and 8.42% in the control group respectively. The frequency of allele was 79.30%, the frequency of T allele was 20.70% and that of control group was 70.30% and 29.70%, respectively. There was significant difference between the two. The CALB index in the CC group (19.019 ± 6.932), CT + TT group (15.835 ± 6.047), the difference was statistically significant (P = 0.002). Conclusion The polymorphism of -735C / T at the promoter region of MMP-2 gene may be related to the susceptibility of viral meningitis. C gene may be a susceptible gene, while T gene may be a protective gene. Polymorphism at this site may affect the susceptibility of viral meningitis by affecting the permeability of BBB.
