目的:探讨中药金安是否能促进顺铂诱导C57BL/6J小鼠Lewis肺癌凋亡。方法:荷Lewis肺癌的C57BL/6J小鼠分为生理盐水组、金安组、顺铂组和金安合用顺铂组,通过检测各组小鼠的体质量变化和抑瘤率,应用流式细胞仪和TUNEL法以及提取组织DNA并电泳等方法观察金安合用顺铂对Lewis肺癌的治疗作用。结果:金安组体质量明显增加,顺铂组体质量先升后降,而金安合用顺铂组体质量则先升后降再回升呈“~”型。各治疗组的抑瘤率分别为40.90%,52.04%和47.83%。流式细胞仪检测各组的凋亡率依次为14.95%,11.68%和14.25%。TUNEL法测定各组的凋亡指数依次是(0.79±1.60)%,(4.00±7.15)%,(5.93±5.96)%,(7.07±10.15)%;DNA提取并电泳在金安合用顺铂组有DNALadder出现。结论:金安能增强顺铂诱导Lewis肺癌的凋亡作用,并能减轻其毒副作用。
AIM: To investigate whether Jin’an could promote cisplatin-induced apoptosis of Lewis lung carcinoma in C57BL / 6J mice. Methods: C57BL / 6J mice bearing Lewis lung cancer were divided into normal saline group, Jin’an group, cisplatin group and Jin’an combined with cisplatin group. By detecting the change of body weight and tumor inhibition rate of mice in each group, Cytometry and TUNEL method as well as the extraction of tissue DNA and electrophoresis and other methods observed Jin’an combined with cisplatin on the treatment of Lewis lung cancer. Results: The body mass of Jin’an group increased significantly, the body mass of cisplatin group increased first and then decreased, while the body mass of Jin’an combined cisplatin group rose first and then dropped back and appeared “~”. The inhibition rate of each treatment group was 40.90%, 52.04% and 47.83% respectively. The apoptotic rate of each group was 14.95%, 11.68% and 14.25% respectively by flow cytometry. TUNEL assay showed that the apoptotic index in each group was (0.79 ± 1.60)%, (4.00 ± 7.15)%, (5.93 ± 5.96)%, (7.07 ± 10.15)%, respectively. DNA extraction and electrophoresis were performed in Jin’an combined with cisplatin DNALadder appears. Conclusion: Jin’an can enhance cisplatin-induced apoptosis in Lewis lung cancer and reduce its side effects.
