目的:测定突变型Nbs1基因转染头颈癌细胞后对放射敏感性的影响。方法:用突变型Nbs1腺病毒载体转染头颈癌细胞株JHU006,同时给予Co60γ射线照射2Gy,于24h后,分别用MTT法测定细胞生长曲线,以及中性彗星法检测癌细胞DNA双链断裂程度。结果:JHU006细胞株在转染突变型Nbs1基因后,再配合放射治疗,癌细胞生长处于停滞状态。中性彗星实验显示,该组癌细胞平均尾力矩值明显增大。结论:突变型Nbs1基因转染可以抑制头颈癌细胞DNA双链断裂损伤的修复功能,因而显著增加了癌细胞放疗敏感性。
Objective: To determine the radiosensitivity of mutant Nbs1 gene transfected into head and neck cancer cells. Methods: The head and neck cancer cell line JHU006 was transfected with the mutant Nbs1 adenovirus vector and irradiated with Co60γ radiation for 2 Gy. The cell growth curve was measured by MTT assay and the degree of DNA double strand breaks . Results: After transfection of mutant Nbs1 gene, JHU006 cells were treated with radiation therapy and the growth of cancer cells was at a standstill. Neutral comet assay showed that the average tail torque of the group of cancer cells increased significantly. CONCLUSION: Transfection of mutant Nbs1 gene can restrain the repair function of DNA double-strand breaks in head and neck cancer cells, and thus significantly increase the radiosensitivity of cancer cells.
