目的观察葱白提取物和丹参酮ⅡA磺酸钠对急性心肌缺血兔血清C-反应蛋白(CRP)、丙二醛(MDA)和超氧化物歧化酶(SOD)的影响,探讨葱白提取物和丹参酮ⅡA磺酸钠治疗急性心肌缺血的作用机制并比较其优势。方法选用体重为2.5~3.0kg的健康雄性日本大耳白兔40只,随机分为假手术组、空白模型组、丹参酮ⅡA磺酸钠、硝酸甘油组和葱白提取物组,每组8只。制备急性心肌缺血模型,术后及时给药,假手术组和模型组舌下给予生理盐水(0.2ml/kg),葱白提取物组舌下给予葱白提取物混悬液(20mg/kg、100mg/ml),硝酸甘油组舌下给予硝酸甘油混悬液(1mg/kg、5mg/ml),丹参酮ⅡA磺酸钠组舌下给予丹参酮ⅡA磺酸钠(1mg/kg、5mg/ml)。给药后观察3h,打开腹腔,腹主动脉取血,测定血清CRP、SOD和MDA含量。结果与模型组比较,葱白提取物组、丹参酮ⅡA磺酸钠组、硝酸甘油组血清中MDA水平均显著降低(P<0.05),而SOD水平均明显升高(P<0.01)。葱白提取物组和丹参酮ⅡA磺酸钠组SOD水平均明显高于硝酸甘油组(P<0.05),葱白提取物组和丹参酮ⅡA磺酸钠组之间SOD水平无统计学差异。葱白提取物组、丹参酮ⅡA磺酸钠组、硝酸甘油组血清中MDA水平比较差异无统计学意义(P>0.05)。葱白提取物组和丹参酮ⅡA磺酸钠组血清CRP水平的降低差异无统计学意义(P>0.05),均明显优于硝酸甘油组(P<0.01)。结论葱白提取物和丹参酮ⅡA磺酸钠均可降低MDA和CRP水平,升高SOD水平,两者作用差异无统计学意义(P>0.05),表明通阳中药葱白有效部位提取物和活血化瘀中药丹参有效成分丹参酮ⅡA磺酸钠均可以清除氧自由基,抗氧化损伤。
Objective To observe the effects of light blue extract and sodium tanshinone Ⅱ A sulfonate on the levels of serum C-reactive protein (CRP), malondialdehyde (MDA) and superoxide dismutase (SOD) in rabbits with acute myocardial ischemia Ⅱ A Sodium Sulfonate Treatment of Acute Myocardial Ischemia Mechanism and to compare their advantages. Methods Forty male Japanese white rabbits weighing 2.5-3.0 kg were randomly divided into sham-operation group, blank model group, sodium tanshinone Ⅱ A sulfonate group, nitroglycerin group and light blue extract group, with 8 rats in each group. The model of acute myocardial ischemia was established. The rats were given sham-operation group and model group subcutaneously with normal saline (0.2ml / kg) / ml). Nitroglycerin was given sublingual nitroglycerin suspension (1mg / kg, 5mg / ml) and tanshinone Ⅱ A sulfonate group was given sublingual tanshinone Ⅱ A sulfonate (1mg / kg, 5mg / ml). After administration for 3h, the abdominal cavity and abdominal aorta were taken for blood and the levels of serum CRP, SOD and MDA were measured. Results Compared with the model group, the serum MDA levels of the extract group, the tanshinone Ⅱ A sulfonate group and the nitroglycerin group were significantly decreased (P <0.05), while the SOD levels were significantly increased (P <0.01). The levels of SOD in scallion extract group and tanshinone ⅡA sodium sulfonate group were significantly higher than those in nitroglycerin group (P <0.05). There was no significant difference in SOD level between scallion extract group and tanshinone Ⅱ A sulfonate group. There was no significant difference in the serum MDA levels between the extract of Allium glabrata, tanshinone Ⅱ A sulfonate group and nitroglycerin group (P> 0.05). There was no significant difference in the serum CRP levels between the extract of scallion extract and sodium tanshinone Ⅱ A sulfonate group (P> 0.05), which was significantly better than that of nitroglycerin group (P <0.01). Conclusion Both the extract of scutellaria and sodium tanshinone ⅡA can reduce the levels of MDA and CRP, and increase the level of SOD. There is no significant difference between the two groups (P> 0.05) The active ingredient of traditional Chinese medicine Salvia tanshinone Ⅱ A sulfonate can scavenge oxygen free radicals, anti-oxidative damage.
