目的观察疏肝饮对内脏高敏感性大鼠结肠组织中TRPA1和PAR2表达的影响。方法 36只Wistar雄性大鼠随机分为正常组、模型组、疏肝饮大剂量组、疏肝饮中剂量组、疏肝饮小剂量组、西药组,采用慢性避水应激法造模10天,造模第4天起,各给药组大鼠于造模前半小时分别给予药物灌胃,连续7天,第10天造模结束后,处死大鼠,取出结肠组织,采用realtime-PCR及免疫组化法分别检测TRPA1和PAR2含量。结果与正常组相比,模型组结肠中TRPA1和PAR2的含量显著增加(P<0.001或P<0.01);与模型组比较,疏肝饮组大、中剂量组和得舒特组大鼠结肠中TRPA1和PAR2含量均明显减少,差异有统计学意义(P<0.001或P<0.01)。结论疏肝饮可能通过调节结肠中TRPA1和PAR2的表达,从而降低内脏高敏感性。
Objective To observe the effect of Shugan Decoction on the expression of TRPAl and PAR2 in the colon tissue of visceral hypersensitivity rats. Methods Thirty-six male Wistar rats were randomly divided into normal group, model group, Shugan Yin large dose group, Shugan Yin medium dose group, Shuganyin low dose group and western medicine group. On the 4th day of model making, the rats in each drug administration group were given gavage for half an hour before the model making for 7 days. After the model was finished on the 10th day, the rats were killed and the colon tissues were removed. Realtime-PCR And immunohistochemistry were detected TRPA1 and PAR2 content. Results Compared with the normal group, the contents of TRPAl and PAR2 in the model group were significantly increased (P <0.001 or P <0.01). Compared with the model group, TRPA1 and PAR2 were significantly decreased, the difference was statistically significant (P <0.001 or P <0.01). Conclusion Shugan Decoction may reduce visceral hypersensitivity by regulating the expression of TRPAl and PAR2 in the colon.
