目的:用雌性大鼠去势建立绝经后骨质疏松症动物模型,去势后分别在白天和晚上进行脉冲电磁场(pulsingelectromagneticfields,PEMF)治疗,观察昼夜节律对PEMF对去势后大鼠骨质疏松症的预防作用的影响。方法:32只雌性SD大鼠随机分为4组,去势组、对照组、去势+PEMF白天预防组和去势+PEMF夜间预防组。各组大鼠分别于术后12周处死,处死前,测量全身骨密度。处死后测定血清骨钙素、行右股骨远端骨形态计量法检查。结果:PEMF白天预防组与PEMF夜间预防组相比BGP降低(1.54±0.11),(1.71±0.15)μg/L,t=2.559,P<0.05、骨小梁间隙减少(127.5±13.3),(143.4±13.9)μm,t=2.337,P<0.05,而全身骨密度增加(0.298±0.010),(0.289±0.008)g/cm2,t=2.283,P<0.05、骨小梁面积百分比增加(35.2±3.0)%vs(32.0±2.4)%,t=2.308,P<0.05、骨小梁宽度增加(65.9±5.2),(59.8±4.9)μm,t=2.435,P<0.05、骨小梁数目增加(5.5±0.4),(5.1±0.3)/mm,t=2.813,P<0.05。结论:PEMF对大鼠卵巢切除引起的骨质疏松症有预防作用,而昼夜节律对PEMF的治疗作用有影响,用PEMF预防大鼠骨质疏松症时白天治疗的效果优于夜间治疗。
OBJECTIVE: To establish an animal model of postmenopausal osteoporosis by castration in female rats. After castration, pulsingelectromagneticfields (PEMF) were performed in both daytime and nighttime to observe the effect of circadian rhythm on PEMF-induced osteoporosis in ovariectomized rats The impact of disease prevention. Methods: Thirty-two female Sprague-Dawley rats were randomly divided into 4 groups: castration group, control group, castrated + PEMF daytime preventive group and castrated + PEMF nighttime preventive group. The rats in each group were sacrificed at 12 weeks after operation, and whole body BMD was measured before sacrifice. Serum osteocalcin was measured after sacrifice and the right femur distal bone morphology was measured. Conclusion: PEMF can prevent osteoporosis induced by ovariectomy in rats, while circadian rhythm has an effect on the therapeutic effect of PEMF. PEMF is superior to nighttime treatment in prevention of osteoporosis.
