目的观察脐血间质干细胞(UCB-MSCs)移植对肌萎缩侧索硬化(ALS)患者血浆及脑脊液谷氨酸(Glu)浓度的影响。方法治疗组8例ALS患者采用静脉滴注结合腰椎脊髓蛛网膜下腔注射UCB-MSCs悬液,行UCB-MSCs移植治疗。应用柱前衍生反相高效液相色谱法分别测定对照组(非神经系统疾病组)和治疗组不同治疗时间血浆和脑脊液中Glu的浓度。结果与治疗前相比,治疗组治疗后3周、3个月血浆、脑脊液Glu浓度降低(P<0.05),但均高于对照组(P<0.05);治疗后3周与3个月相比,治疗组血浆、脑脊液Glu浓度差异无统计学意义(P>0.05)。结论脐血间质干细胞移植治疗能够降低ALS患者血浆、脑脊液中Glu浓度。
Objective To observe the effects of umbilical cord blood mesenchymal stem cells (UCB-MSCs) transplantation on glutamate (Glu) concentration in plasma and cerebrospinal fluid of amyotrophic lateral sclerosis (ALS) patients. Methods Eight patients with ALS were treated with UCB-MSCs transplantation by intravenous drip combined with lumbar spinal subarachnoid injection of UCB-MSCs. Pre-column derivatization reversed-phase high performance liquid chromatography was used to determine the concentration of Glu in plasma and cerebrospinal fluid in control group (non-neurological disease group) and treatment group at different treatment time. Results Compared with those before treatment, the concentration of Glu in plasma and cerebrospinal fluid in the treatment group decreased 3 weeks and 3 months after treatment (P <0.05), but both were higher than those in the control group (P <0.05). After 3 weeks and 3 months There was no significant difference in Glu concentration in plasma and cerebrospinal fluid between the two groups (P> 0.05). Conclusion Umbilical cord blood mesenchymal stem cell transplantation can reduce the concentration of Glu in plasma and cerebrospinal fluid of patients with ALS.
