目的:研究短瓣金莲花总黄酮的大孔树脂纯化工艺条件。方法:通过对短瓣金莲花总黄酮在3种大孔吸附树脂上的吸附与解吸附特性的比较,筛选最佳树脂,并对该树脂的吸附特性及各影响因素进行系统研究,采用正交试验及单因素考察等方法对纯化工艺条件进行优选。结果:以D101大孔吸附树脂为分离载体,样品液浓度为0.5 g/mL,pH6.0,上样量为药材∶树脂=1∶1(W/W),上样后静态吸附1 h,先用4 BV水洗脱除杂,再用6 BV 30%乙醇解吸,所得产品中总黄酮平均含量为60%以上。
Objective: To study the purification conditions of macroporous resin of total flavonoids from Trollius chinensis. Methods: The best resin was selected by comparing the adsorption and desorption properties of total flavonoids of T. cuspidatum on three kinds of macroporous adsorption resins. The adsorption characteristics and various influencing factors of the resin were systematically studied. Tests and single-factor investigations and other methods optimize the purification process conditions. Results: The D101 macroporous adsorption resin was used as the carrier. The concentration of the sample solution was 0.5 g/mL, pH was 6.0, and the sample amount was drug:resin=1:1(W/W). The sample was statically adsorbed for 1 h. First elute with 4 BV water to remove impurity, and then desorb with 6 BV 30% ethanol. The average content of total flavonoids in the obtained product is more than 60%.
