目的探讨共患和不共患破坏性行为障碍(DBD)的儿童注意缺陷多动障碍(ADHD)与5-HT2C 受体基因(HTR2C)C-759T 和 G-697C 两种多态之间的关联。方法就237个共患 DBD的 ADHD 核心家系和251个不共患 DBD 的 ADHD 核心家系的 C-759T 和 G-697C 两种多态进行检测,并对各等位基因和单体型进行传递不平衡检验(TDT)。结果在共患 DBD 的家系中未发现任何等位基因和单体型的传递不平衡现象。在不共患 DBD 的家系中,-759C(X~2=4.25,P=0.04)优先传递,-697G(X~2=3.21,P=0.07)有优先传递的趋势,就单体型而言,-759C/-697G 优先传递(X~2=4.31,P=0.04)。结论 ADHD 共患 DBD 与否在 HTR2C 基因 C-759T 和 G-697C 两种多态水平上有明显的遗传差别。HTR2C 基因与不共患 DBD 的 ADHD 相关联。
Objective To investigate the association between children with ADHD and 5-HT2C receptor gene (HTR2C) C-759T and G-697C polymorphisms in children with and without comorbid destructive behavior disorder (DBD) . METHODS: Two polymorphisms of C-759T and G-697C in 237 core families of ADHD with comorbid DBD and 251 core families of ADHD with no DBD were detected and alleles and haplotypes were not transmitted Balance test (TDT). Results No allelic and haplotypic transmission imbalance was found in the pedigrees of comorbid DBD. In pedigrees with no common DBD, -759C (X ~ 2 = 4.25, P = 0.04) was preferentially transmitted and -697G (X ~ 2 = 3.21, P = 0.07) had a preferential transmission. In terms of haplotype , -759C / -697G preferentially (X ~ 2 = 4.31, P = 0.04). Conclusion ADHD comorbid DBD or not in the HTR2C gene C-759T and G-697C two polymorphic levels have significant genetic differences. The HTR2C gene is associated with ADHD without DBD.
