本文报道用人胃癌(粘液腺癌Ⅱ级)组织提取液免疫的Balb/c小鼠脾细胞和SP2/0小鼠骨髓瘤细胞融合,获得一个抗人IgA单克隆抗体(简称McAb),命名为A_5B_8,IgG类属IgG_3。腹水抗体滴度为1:10~(-5)~1:10~(-6)。竞争抑制试验显示,人IgG对McAb无抑制作用,而人初乳SIgA、胃癌组织和癌旁胃粘膜组织中SIgA对反应有明显抑制作用,证明McAb是抗人IgA抗体。但是McAb对不同来源的IgA的亲和性不同,如人初乳SIgA浓度为0.5mg/ml、1.0mg/ml和2.0mg/ml时,其抑制率分别为60.3%、61.6%和65.8%增加不明显。而胃癌组织SIgA同样3种浓度,其抑制率分别为39.4%、69.8%和80.5%,抑制作用明显增加。癌旁胃粘膜SIgA的抑制作用与胃癌SIgA相同,分别
This article reports the fusion of Balb/c mouse spleen cells immunized with human gastric cancer (mucinous adenocarcinoma grade II) tissue extract and SP2/0 mouse myeloma cells to obtain an anti-human IgA monoclonal antibody (abbreviated as McAb), named A_5B_8 IgG class is IgG_3. Ascites antibody titer was 1:10~(-5)~1:10~(-6). The competitive inhibition test showed that human IgG had no inhibitory effect on McAb, but SIgA in human colostrum SIgA, gastric cancer tissues, and adjacent gastric mucosal tissues significantly inhibited the reaction, demonstrating that McAb is an anti-human IgA antibody. However, the affinity of McAb for different sources of IgA is different. For example, when the concentrations of human colostrum SIgA are 0.5 mg/ml, 1.0 mg/ml and 2.0 mg/ml, the inhibition rates are 60.3%, 61.6% and 65.8%, respectively. Not obvious. The same three concentrations of gastric cancer tissue SIgA, the inhibition rate were 39.4%, 69.8% and 80.5%, significantly increased inhibition.
