甲状腺相关眼病眼眶组织透明质酸酶mRNA表达的研究

目的探讨透明质酸酶1(HYAL1)mRNA在甲状腺相关眼病(TAO)球后脂肪组织及眼外肌中的表达情况,为透明质酸酶用于TAO患者的治疗提供实验依据。方法收集我院2000~2005年确诊并行眼眶减压术或斜视矫正术的TAO患者共21例为TAO组,获得眼外肌标本18例,脂肪标本17例;正常肌肉组织标本17例和脂肪组织标本11例作为对照组。所有标本通过原位杂交的方法,测量累计光密度(IOD)值均值,比较HYAL1mRNA的表达。结果 TAO肌肉组织中HYAL1 IOD均值高于对照组(P<0.01);而TAO和对照组脂肪组织中HYAL1 IOD均值比较差异无统计学意义。结论 TAO患者肌肉组织中有高的HYAL1 mRNA表达量,可能与眼外肌的纤维化有关;脂肪组织中有低水平的HYAL1 mRNA表达,是造成眶后透明质酸堆积的原因之一。 Objective To investigate the expression of hyaluronidase 1 (HYAL1) mRNA in the adipose tissue and extraocular muscles of patients with thyroid associated ophthalmopathy (TAO) and provide experimental evidence for the treatment of TAO with hyaluronidase. Methods Twenty-one patients with TAO diagnosed concurrent orbital decompression or strabismus surgery in our hospital from 2000 to 2005 were selected as TAO group. Eighteen cases of extraocular muscle samples and 17 cases of fat samples were obtained. 17 cases of normal muscle tissue samples and 18 cases of adipose tissue Eleven specimens were used as control group. All specimens by in situ hybridization method, measuring the mean optical density (IOD) values, compared HYAL1 mRNA expression. Results The mean IALOD of HYAL1 in TAO muscle tissue was higher than that in control group (P <0.01). There was no significant difference in mean IALOD of HYAL1 between TAO and control group. Conclusions The high expression of HYAL1 mRNA in muscle tissue of TAO patients may be related to the fibrosis of extraocular muscles. The low level of HYAL1 mRNA expression in adipose tissue is one of the reasons for the accumulation of hyaluronic acid in the orbit.
