目的 了解不同浓度的瘦素 ,在不同条件下对离体大鼠胰岛分泌胰岛素的影响。方法利用细胞培养技术 ,在不同的葡萄糖浓度 (5 .6mmol/L或 16 .7mmol/L)和不同作用时间 (10min或2h)下 ,以 0 ,1,5 ,10 ,15 ,5 0或 10 0 μg/L瘦素作用于大鼠胰岛 ,观察其对胰岛素分泌的影响 ;用放免法检测培养物上清液胰岛素浓度。结果 培养液葡萄糖浓度为 5 .6mmol/L ,培养 10min ,1μg/L、5 μg/L瘦素促进被孵育的胰岛的胰岛素分泌 ;培养 2h ,5 μg/L瘦素促进基础胰岛素分泌 ,≥ 5 0 μg/L瘦素抑制胰岛素分泌。葡萄糖浓度为 16 .7mmol/L ,培养 10min ,≥ 5 0 μg/L瘦素抑制胰岛素分泌 ;培养2h ,≥ 5 μg/L瘦素抑制胰岛素分泌。 结论 重组瘦素对离体大鼠胰岛分泌胰岛素具有双向作用 ,并受瘦素浓度、作用时间和环境葡萄糖浓度等因素变化的影响
Objective To investigate the effect of different concentrations of leptin on insulin secretion in isolated rat islets under different conditions. Methods Using cell culture techniques, cells were cultured at 0, 1, 5, 10, 15, 50 or 10 at different glucose concentrations (5.6 mmol / L or 16.7 mmol / L) and different duration of action (10 min or 2 h) The effect of leptin at 0 μg / L on rat islets was observed and the insulin secretion of the supernatant was detected by radioimmunoassay. Results The concentration of glucose in culture medium was 5.6 mmol / L, and the leptin of 1 μg / L, 5 μg / L was added to promote the insulin secretion of the incubated islets. After cultured for 2 h, 5 μg / L leptin promoted basal insulin secretion, ≥ 5 Leptin at 0 μg / L inhibits insulin secretion. Glucose concentration of 16.7mmol / L, cultured for 10min, ≥ 50μg / L leptin inhibited insulin secretion; cultured 2h, ≥ 5μg / L leptin inhibited insulin secretion. Conclusions Recombinant leptin has a bidirectional effect on the insulin secretion in isolated rat islets and is influenced by such factors as leptin concentration, duration of action, and environmental glucose concentration
