目的:建立超高效液相色谱法(UPLC)同时测定穿心莲内酯磺化物中穿心莲内酯磺化物E、穿心莲内酯磺化物F含量。方法:色谱柱:Shimadzu Shim-pack XR-ODSⅢ(2.0 m×75 mm,1.6μm);流动相:乙睛(A),磷酸二氢钾缓冲液(B:每1000 ml水中加入磷酸二氢钾1.361 g与庚烷-1-磺酸钠1.0 g)梯度洗脱;流速:0.4 ml·min-1;检测波长:225 nm;柱温:30℃。结果:穿心莲内酯磺化物E在4.39~43.88 g·ml-1(r=1.000 0)、穿心莲内酯磺化物F在4.43~44.32μg·ml-1(r=1.000 0)范围内,呈良好的线性关系。穿心莲内酯磺化物E平均回收率为98.71%,RSD=1.41%(n=6);穿心莲内酯磺化物F平均回收率为99.29%,RSD=0.76%(n=6)。结论:本法重复性好、灵敏度高、定量准确,可用于穿心莲内酯磺化物的质量控制。
OBJECTIVE: To establish a method for simultaneous determination of andrographolide S and andrographolide F in andrographolide sulphonate by ultra performance liquid chromatography (UPLC). Methods: Column: Shimadzu Shim-pack XR-ODSⅢ (2.0 m × 75 mm, 1.6 μm); mobile phase: acetonitrile (A), potassium dihydrogen phosphate buffer 1.361 g and heptane-1-sulfonate 1.0 g); flow rate: 0.4 ml · min -1; detection wavelength: 225 nm; column temperature: 30 ° C. RESULTS: The concentrations of andrographolide sulfonate E ranged from 4.39 to 43.88 g · ml-1 (r = 1.000 0) and the concentrations of andrographolide sulfonate F ranged from 4.43 to 44.32 μg · ml-1 (r = 1.000 0) The linear relationship. The average recovery of andrographolide sulfonate E was 98.71%, RSD = 1.41% (n = 6). The average recovery of andrographolide S was 99.29% and RSD was 0.76% (n = 6). Conclusion: This method has good repeatability, high sensitivity and quantitative accuracy, which can be used for quality control of andrographolide sulfonate.
