目的进一步明确环氧化酶-2(COX-2)与前列腺不同病变组织的关系,探讨COX-2对前列腺癌生物学行为的影响;方法采用免疫组织化学方法对正常前列腺、良性前列腺增生、前列腺癌组织的石蜡包埋切片进行染色,检测COX-2蛋白质表达,RT-PCR检测COX-2mRNA表达,分析其与前列腺及其不同病变组织的关系。结果(1)COX-2在正常前列腺组织中表达为阴性,良性前列腺增生、前列腺癌组织中均见表达,1染色的阳性系数均值分别为0.250±0.500,1.500±0.972,2.333±1.231,前列腺癌组织中表达程度较高。(2)COX-2mRNA在正常前列腺、良性前列腺增生及前列腺癌组织中表达的相对光密度值分别为0.053±0.062,0.589±0.161,0.833±0.103,差异有统计学意义。结论COX-2在前列腺癌的发生发展中起重要作用并影响前列腺癌的生物学行为,COX-2可能成为前列腺癌治疗的靶分子。
Objective To further clarify the relationship between cyclooxygenase-2 (COX-2) and different pathological changes of prostate and to explore the effect of COX-2 on the biological behavior of prostate cancer. Methods Immunohistochemistry was used to detect the expression of COX-2 in benign prostatic hyperplasia The expression of COX-2 protein was detected by immunohistochemical staining in paraffin-embedded sections of cancer tissues. The expression of COX-2 mRNA was detected by RT-PCR and the relationship between the COX-2 mRNA and the prostate and its different pathological tissues. Results (1) The expression of COX-2 in normal prostate tissue was negative, benign prostatic hyperplasia and prostate cancer tissues, respectively. The mean values of positive staining for 1 staining were 0.250 ± 0.500 and 1.500 ± 0 respectively. 972,2.333 ± 1.231, higher expression in prostate cancer tissues. (2) The relative optical density of COX-2 mRNA in normal prostate, benign prostatic hyperplasia and prostate cancer tissues were 0.053 ± 0.062,0.589 ± 0.161,0.833 ± 0.103, The difference was statistically significant. Conclusions COX-2 plays an important role in the development of prostate cancer and affects the biological behavior of prostate cancer. COX-2 may be the target of prostate cancer treatment.
